Chalmers, 1964] were diluted with warmed TC 199 culture medium (Glaxo) in sterile siliconed universals with white rubber liners, and then cultured. These cultures were made from the blood of the same healthy donors who provided the fresh small lymphocytes. These donors, A.C., D.W., and L.F. had previously had prophylactic tetanus toxoid injections. The tissue culture cell concentration was adjusted to 1000 small lymphocytes/cu.mm. Adsorbed tetanus toxoid, Tet/Vac/ADS (Burroughs Wellcome), was used as the antigenic stimulus; 041 ml. of the toxoid was added per 10 ml. of culture medium. Previous experience indicated that this concentration of antigen results in an optimal antigen to cell ratio.
