Penelitian ini bertujuan melihat hubungan dan kontribusi antara tingkat pengetahuan lingkungan hidup dengan sikap peduli lingkungan siswa di SMA Negeri 14 Medan. Penelitian ini dilakukan terhadap seluruh siswa kelas X yang terdiri dari 10 kelas, dengan jumlah populasi 360 orang dan sampel 130 orang siswa. Jenis penelitian ini adalah penelitian deskriptif dengan teknik korelasi. Teknik pengampilan sampel dilakukan dengan cara Random Sampling. Instrumen penelitian yang digunakan adalah tes pengetahuan lingkungan, angket sikap peduli lingkungan, lembar observasi dan wawancara. Hasil Penelitian menunjukkan bahwa nilai korelasi yang terbentuk dari tingkat pengetahuan lingkungan hidup siswa dengan sikap peduli lingkungan siswa adalah sebesar 0,327 (rendah) dengan kontribusi sebesar 10,7 % dan analisis regresi menunjukkan persamaan regresi Ŷ = 81,557 + 0,157 X. Dari hasil perhitungan diperoleh thitung 3,915 > ttabel 1,9978. Hal ini mengindikasikan bahwa Ha diterima dan H0 ditolak, dengan kata lain ada hubungan yang signifikan antara tingkat pengetahuan lingkungan hidup dengan sikap peduli lingkungan siswa di SMA Negeri 14 Medan.
White leafhopper Cofana spectra is a pest of paddy and could act as a vector of pathogenic viruses on rice. This research aims to find out its genetic data, namely its identification, nucleotide composition and its phylogenetic based on DNA barcode-mtCOI gene. The researchmethods included leafhopper sampling in rice field, DNA isolation, PCR, electrophoresis, sequencing and blasting. The software Mega X was used to analyze the genetic data. The mtCOI DNA gene sequence obtained was 333 bp in size and blasting result showed that leafhopper belongto white leafhopper Cofana spectra (Distant) species. The dendrogram from the sequence showed that white leafhopper species from Samosir island had the closest relationship with species from Kerala, India. The frequency of nucleotide of T (U), C, A and G of leafhopper were 41.96, 15.47, 28.57 and 13.98%, respectively. Therefore, this mtCOI gene sequence was rich in A/T (70.53%), while G/C was 29.45%. The information obtained from this study stated that the identification of the white leafhopper C.spectra based on molecular character using mtCOI DNA gene sequence confirmed the identification result based on the morphological traits and can be used to control and in monitoring the spread of the white leafhopper.
The zigzag leafhopper Maiestas dorsalis (Hemiptera) is a tungro virus vector that cause damage on rice plants. Genetic analysis of M. dorsalis that be isolated from Samosir island, North Sumatra, Indonesia using partial DNA sequence of mitochondrial cytochrome oxidase subunit I (COI) DNA is still limited. This study aims to identify and to find out the genetic data of M. dorsalis that be derived from region of its mitochondrial cytochrome oxidase I gene (COI). Analysis was done by four steps, namely: Hopper collection on paddy field by using insect net, DNA extraction by using Zymo Tissue & Insect DNA Mini Preparation, amplification by PCR using My TaqTM HS Red Mix and DNA sequence analysis using ABI PRISM 3730xl Genetic Analyzer. Primer cocktail tRWF-Mlep was used in DNA amplification step. The research result pointed out that COI DNA fragment of M. dorsalis has length 521 bp. This COI DNA sequence was dominated by A and T(U) bases with concentration 74.30%. The concentration of T(U), C, A and G nucleotides in the COI sequence were 35.90%, 13.40%, 38.44%, and 12.30%, respectively. Identification of M. dorsalis based on this COI DNA sequence confirmed the identification result based on its morphological characters.
