Context Salt stress harms plant growth and limits crop productivity. Autophagy is a material decomposition and metabolism pathway that is central to regulating plant responses to environmental stimuli and may be employed in plant breeding for stress tolerance. The autophagy-related gene ATG8 has become an essential tool for monitoring autophagic activity. Aims We aimed to determine the effects of increased autophagic activity via overexpression of ATG8 on tolerance of wheat (Triticum aestivum L.) seedlings to NaCl stress. Methods Salt-tolerant wheat cv. Jimai22 was used to produce TaATG8a-overexpressing wheat seedlings using Foxtail mosaic virus based virus-mediated overexpression technology. TaATG8a-overexpressing seedlings were exposed to NaCl stress for 6 days. Key results Overexpression of TaATG8a enhanced autophagic activity and improved salt stress tolerance of wheat seedlings. Overexpression of TaATG8a also reduced damage to photosystem II, reduced excess accumulation of reactive oxygen species, increased the activities of superoxide dismutase, peroxidase and catalase, and suppressed programmed cell death in wheat seedling leaves during NaCl treatment. Conclusions Overexpression of TaATG8a conferred higher tolerance of wheat seedlings to NaCl stress and suppressed salt-induced programmed cell death, accompanied by increased autophagic activity. Implications This study provides genetic resources and a theoretical basis for stress tolerance breeding in wheat.
Objective: To test the reliability between two instruments with different analysis mechanisms, either by GPS (model GPSPORTS®) or by video analysis (InStat For Players®), relating the results of total distance covered and distance at high speed ≥ 20km/h (Very High-Intensity Running Distance, VHIR) during official soccer matches. Study Design: This is a methodological study. Data from 35 male professional soccer athletes from all tactical positions were included. Age 29.2 (± 4.8 years) and body fat 9.9 (± 1.7%), excluding goalkeepers (102 individual analyzes) were collected in official matches. In the data analysis, descriptive statistics procedures were used to characterize the sample and the intraclass correlation coefficient (ICC) was used to verify the agreement on the stability and internal consistency of the tests with 95% confidence intervals (CI). Results: The ICC in the case of the total distance traveled variable was significant 0,914 (0,876; 0,941) and indicated a very high agreement, with the linear correlation coefficient indicating a strong positive correlation (p <0.001). The ICC for the VHIR variable was not significant, although the linear correlation coefficient indicates a strong positive correlation (p <0.001). Clinical Relevance Statment: This study reveals that there is good agreement in the comparison of two systems designed to analyze the movement demands of each professional soccer athlete in relation to the total distance covered. Introduction: Exercise commitment plays an explanatory and guiding role in an individual's adherence to exercise behavior. However, exercise commitment in recreational situations is poorly studied. Objective: Develop a sporting and recreational behavior questionnaire based on the commitment scale for Chinese urban residents. Methods: Two stages were conducted, the first with a sample of 453 individuals to explore and verify the determinants of behavioral commitment in recreational sport and the fit of the framework, and the second with a sample of 786 individuals to investigate the relationship between behavioral commitment in recreational sport and its determinants. Results: Behavioral commitment in-play sport included five determinants: ‘motivation to participate’, ‘personal involvement’, ‘choice to participate’, ‘opportunity to participate’, ‘social support’. The constructed model showed a good fit. The factors: ‘motivation to participate’, ‘personal engagement’ and ‘interest to participate’, showed a significant positive association with engagement and a significant negative association with a choice to participate. Conclusion: The resulting model shows characteristics of recreational sport behavior and can be used as a measure of commitment to recreational sport behavior among residents of a Chinese city.
