BACKGROUND Leprosy is an infectious disease that is still a health problem worldwide, including in Indonesia. The clinical symptoms are similar to other skin diseases and it is difficult to establish a diagnosis for paucibacillary (PB) leprosy. Current serological and histopathological tests have limitations, especially in patients with negative acid-fast bacilli (AFB). Serological tests often give false-negative results, while histopathological results often consist of non-specific inflammation. Probe-based real-time polymerase chain reaction (RT-PCR) assays is an alternative test that may be more sensitive and more specific to detect Mycobacterium leprae.METHODS This study was done in June 2015 until March 2016; detected M. leprae in PB patients with negative AFB smears using TaqMan® probe-based RT-PCR assay on slit skin scrapings and skin biopsy specimens from 24 patients. The skin scrapings were obtained from skin tissue on ear lobes, skin lesions, as well as those from biopsy. Samples were tested with RT-PCR while histopathological examinations were only performed on skin from biopsy. RESULTSThe RT-PCR assay showed positive results of 21%, 25%, and 96% for specimens obtained from skin scrapings of the ear lobe, skin lesions, and skin biopsy, respectively. On the other hand, the positive rate for the histopathological test from skin biopsy was 79%. It indicated that the TaqMan® RT-PCR assay could increase the diagnostic capacity of histopathological examination by as much as 17%.CONCLUSIONS TaqMan® PCR assay can improve the diagnostic capacity of histopathological examinations, which could be used as the new gold standard for the diagnosis of leprosy.
Currently, obesity is a problem for both developed and developing countries. Obesity nutritional status is associated with body fat levels, central obesity and is associated with various diseases This study analyzes the correlation between body mass index, waist circumference, and the ratio of waist to hip to body fat percentage and visceral fat based on gender. An observational analytic study with a cross-sectional design, involving sixty-three employees aged 19-60 years in February 2021. Subject characteristics data were collected using a questionnaire, in addition to physical examination to obtain anthropometric data in the form of weight, height, waist circumference, hip circumference. Data on the percentage of body fat and visceral fat were obtained using the Mi body composition digital tool. The Pearson's correlation test was used for analysis and the significance level used was 0.05. A total of 49.2 % of subjects were categorized as obese based on body mass index. In the male group, there was a strong and significant positive correlation between BMI, WC, with the percentage of body fat and visceral fat with p =0.000. However, in female subjects, a significant positive correlation was found between BMI, WC, with visceral fat in women with p = 0.000. BMI, WC, and WHR correlated strongly with body fat and visceral fat levels in men. In the female group, BMI and WC were strongly correlated with visceral fat. Keywords: anthropometric, fat, visceral
Dengue hemorhagic fever (DHF) is an infection caused by dengue virus. Dengue is a virus that is transmitted from Aedes Spp mosquito, the fastest growing mosquito in the world and has transmitted dengue virus. The prevalence of DHF until July 2020 has reached 71.633 in Indonesia, and 4.227 cases only in DKI Jakarta province. We usually don’t find new case on July, but this year we still find new cases on July. This should increase our vigilance, especially at this time when there is a pandemic of COVID-19 cases. The purpose of this activity is to increase public knowledge about dengue fever so that they can prevent dengue fever in their area. This activity is carried out through seminar using the zoom application because conditions in the field do not allow offline activities to be held. The activity will be held on Saturday, April 23, 2022. The results obtained through this PKM activity is an increase in the level of participants' knowledge about DHF as evidenced by an increase in post-test scores after attending the seminar. The conclusion of this PKM activity went well because of the support from all related parties from institutions, solid teams, and partners who actively participated. The shortage of this PKM activity is that the activities are carried out online because the conditions are still not conducive to do an offline activity. The advice we can give is to carry out PKM activities again with the same partner with different health topics according to the conditions that exist in the partners.
Rawa Buaya Village is one of the villages in the West Jakarta. This village is a densely populated village with a limited number of health facilities. Based on the 2018 DKI National Basic Health Research (RISKESDAS) data, the proportion of the population aged over 10 years in West Jakarta who behaved properly in washing their hands was only 49.11%. Hand washing is the most important basic technique in the prevention and control of infection transmission because it mechanically removes dirt and dust from the skin surface and reduces the number of pathogenic microorganisms. Because knowledge of clean and healthy living behavior (PHBS) by washing hands with soap is a determining factor in the transmission of infectious diseases such as diarrhea, respiratory infections including Covid-19, education is needed to increase this knowledge. Education by counseling and training activities are carried out online through the Zoom teleconference on April 23, 2022, followed by 45 village residents. Participants' knowledge was assessed by pre-test and post-test using Google Form. From the post-test’s results, there is an increase in the participants’s knowledge by 22.9% with an increase in the percentage of correct answers from the pre and post test ranging from 6.6% - 35.6%, which is also expected to change in habits or behavior.
