Tobacco leaf disc explants were inoculated with Agrobacterum tumefaciens strain GV2260 carrying p35S GUS-INT to determine the influence of different co-cultivation temperatures (18 -26 °C), periods (24 -96 h) and media (solid and liquid) on transformation efficiency. Kanamycin-resistant shoots developed on leaf discs inoculated with Agrobacterium after 4 weeks of culture initiation. Regenerated shoots were excised and rooted in the basal medium supplemented with 100 mg dm -3 kanamycin and 250 mg dm -3 augmentin. The rooted plantlets were finally transferred to compost and confirmed by GUS assay and PCR analysis. The highest transformation frequency was achieved from the explants cocultivated with A. tumefaciens in liquid medium for 48 h at 22 or 24 °C.
This research was conducted to determine the dry matter yields and some quality features of alfalfa cultivars in Ankara and Isparta locations of Turkey. In this study, six alfalfa cultivars, (Daisy, Prosementi, Savas, Bilensoy, Gea) which one of them is candidate were tested in the randomized block design with three replications in 2007-2009 growing seasons. According to the results, significant differences were determined in terms of plant height, main stem diameter, stem number, dry matter yield, crude protein ratio, ADF and NDF contents of alfalfa cultivars. The highest plant heights were determined from Candidate and Prosementi in Ankara, Candidate, Bilensoy and Prosementi in Isparta. The highest stem numbers were determined in Prosementi, Bilensoy and Candidate in Isparta location. Daisy and Candidate had the highest crude protein content and they had the lowest NDF and ADF contents in both locations of the study. While the highest dry matter yield was obtained from Candidate in Ankara, it was obtained from Candidate and Daisy in Isparta. At the end of the study, for the high dry matter yield and quality candidate cultivar is more suitable for Ankara, while candidate and daisy cultivars are more suitable for Isparta location.
An efficient and reliable micropropagation system for Persian clover (Trifolium resupinatum L.) was developed using different explants and media. Node, hypocotyl and cotyledonary node explants were cultured on Murashige and Skoog (MS) medium supplemented with combinations of either 6-benzyladenine (BA) and indole-3-butyric acid (IBA) or BA, Kinetin (KIN) and IBA. Direct multiple shoots developed within 6 weeks in all explants in most media tested. The best shoot multiplication capacity was obtained from cotyledonary node explants on MS medium containing 7.
