Aims: Defensins are cysteine-rich antimicrobial cationic peptides and BAX is a proapoptotic gene that can cause cell death. This study aimed to investigate the effect of cellular extract of co-cultured Lactobacillus casei on the expression of BAX and human β-defensin 2 (hBD-2) genes in HT29 cells. Methods & Materials: This experimental study was conducted in the Research Center for Pharmaceutical Nanotechnology of Tabriz University of Medical Sciences in 2017. The HT29 cell line was obtained from the Pasteur Institute of Iran, and cells were assessed using Microculture Tetrazolium Test (MTT) after culturing. DNA was extracted from the treated cells, and then the DNA ladder assay was carried out. After preparing cDNA, the expression levels of BAX and hBD-2 genes in the HT29 cell line were measured using a real-time Polymerase Chain Reaction (PCR) method. Findings: The results of the MTT assay indicated that Lactobacillus casei inhibited the proliferation of HT29 cells and induced apoptosis in these cells. Results of DAPI staining and DNA ladder assay obtained from treating HT29 cells by Lactobacillus casei showed qualitative changes in cell apoptosis. Moreover, realtime PCR results indicated that Lactobacillus casei bacteria significantly increased the expression of the hBD-2 gene in HT29 colon cancer cells within 12-24 hours (P= 0.023), while BAX gene expression showed no significant change in the first 24 hours (P= 0.37). Conclusion: The extract of Lactobacillus casei can be used to stimulate cancer cells to produce β-defensins, inhibit pathogens, prevent the stimulation of cellular signaling, and fight antibiotic-resistant bacteria.
Background: Cervical and breast cancer are considered to be the most common cause of death among Iranian women. Various studies have shown that probiotics are effective fighting cancer. The aim of the present study was to investigate the effect of Lactobacillus brevis on apoptosis and casp (casp3, casp8) gene expression in HeLa cancer cells. Materials & Methods: In this study, L. brevis bacteria were collected from the vaginal fluid of females referred to Alzahra Hospital and isolated and cultured in MRS agar medium. Inhibition of HeLa cancer cell proliferation by bacteria was evaluated by MTT assay. Apoptosis of cancer cells was measured by fluorescent microscopy using DAPI method. Finally, expression of Caspase and akt genes was measured by Real time PCR. Results: The results indicated that L. brevis had the same effect on HeLa cancer cells (P=0.42, P=0.26). On the other hand, there was no significant increase in akt gene expression (p> 0.05). Conclusion: Lactobacillus brevis bacteria can be considered biologically safe for the development of a novel, high-impact, low-throughput therapeutic strategy. On the other hand, side-by-side treatment and prevention against cancer will be costeffective.
Background. If the pregnancy attempt fails, it may become a stressful event. Therefore, in many cases, problem solving can lead to greater satisfaction and cohesion of couples and prevent psychological and social harm to families. The aim of this study was to determine the significant difference in the expression of miR-146a miRNA in maternal blood between the control and observation groups and to determine whether the miRNA can be used as diagnostic biomarkers or not. Methods. The present study is a basic and descriptive survey in terms of data analysis. The samples included pregnant women referred to Tabriz University of Jahad from September 2019 to February 2021. We used Exel, Spss v.21 and Roc software for data analysis. Data were prepared in Exel software with reference gene and by calculating CT. Were used in Roc software. Results. The results showed a difference between the control and observation groups. Early detection of recurrent miscarriage should be used in these mothers. Conclusion. Through using diagnostic kits of the studied miRNAs, effective steps can be taken for early detection, prevention of abortion, and reducing the heavy psychological, economic, and social burden on families. Therefore, it is suggested that diagnostic kits be made for these biomarkers and used in infertility centers. Practical Implications. Among the practical implications of this study, we can mention the diagnosis and prevention of abortion, which can reduce the burden on families and society.
