Previous report showed the high potent antiproliferative effect of the methanolic part extracted from the aerial parts of Pouzolzia indica on NB4 and HT93A acute leukemic cell lines with the IC50 values of 28.5 and 49.8 μg/mL, respectively. The bioassay-guided fractionation of the methanolic part gave 5 fractions, that is, FFI–FFV. FFII, FFIII, and FFIV inhibited the above leukemic cell lines with the IC50 values of 15.1 (FFII), 14.4 (FFIII), 32.1 (FFIV), and 31.0 (FFII), 9.7 (FFIII), 10.5 (FFIV) μg/mL, respectively. The compounds in these fractions were isolated using chromatographic technique. FFII contained friedelin 1, 28-hydroxy-3-friedelanone 2, and 7-methoxy-coumarin 3. FFIII contained 6, 7-dimethoxy-coumarin 4, scopoletin 5, methyl caffeate 6. FFIV contained sitosteryl glucoside 7 and a supposed glycosphingolipid 8. The chemical structures were elucidated by spectroscopic methods.
Garcinia speciosa Wall.is Thai medicinal plant used as remedies for healing skin infections and wounds, a laxative (bark), inflammation, astringent (leaves) agents, diarrhoea (fruits) and antibacterial.This plants were rich source of oxygenated xanthones, some of which have antitumor promotive activity. Dried leaves and twigs of G. speciosa extracted with ethanol and chromatography with Diaion20 column by H 2 O, methanol and ethyl acetate gave 3 fractions. The crude extracted of H 2 O, methanol and ethyl acetate fractions were tested by cytotoxic against to the lung cancer cell lines (A 549) gave ED 50 values of 62, 45 and 75μg/ml, respectively. Hence, the crude methanol fraction was interested for chemical constituents and identified cytotoxic activities. Crude methanol fraction was fractionated by si gel column, used of ethyl acetate-hexane (7:3) gave macluraxanthone was major compound. Structure elucidated by 1H, 13C and 2D NMR spectroscopy. The antioxidant of macluraxanthone by DPPH showed IC 50 was 7.65μg/ml and the cytotoxic test of lung cancer cell lines (A 549) gave ED 50 values 15.38 μg/ml.
Acanthamoeba spp. is the free living protozoain the environment which are opportunistic pathogens of human [1]. Acanthamoeba spp. is the causative agents of Acanthamoeba keratitis (AK)[2]. The risks of AK increased by using contact lens are not good hygiene. Antiamoebic therapy were biguanides chlorhexidine (BCH) or polyhexamethylene biguanide (PHMB) which were not effective enough to kill Acanthamoeba cysts [3]. Reported, plant for protected glaucoma, anti-inflammatory and analgesic properties are Peperomiapellucida in Piperaceae family. This interesting of studied P. pellucida for antiamoebic. Dried plant of P. pellucida was chromatographic by Diaion HP20 and fractionated into methanol : water (1:1, FrI), methanol (FrII) and ethyl acetate (FrIII). The methanol fraction (FrII) was damaged morphological and changed structural of Acanthamoeba cysts, detected by toluidine dye staining are shown under the light microscope, in IC50 29.28±3.64%.The active methanol fraction was studied by chromatography, gave four fractionated fraction A1-A4. A1 led to the isolation of compounds 1, A2 and A3 isolated compounds 2-5. They were identified using spectroscopic methods.
