Video-based walking track systems have been developed for gait analysis in rat models. However, there is no previous study using video-based tracking systems to address the gait parameters to evaluate the recovery of Achilles tendon rupture models. This study conducted a comprehensive gait analysis using a video-based image processing system. Eighteen Sprague-Dawley rats were randomly assigned to one of three interventional conditions: sham surgery, Achilles tendon repair, and Achilles tendon defect. After surgery, all animals were evaluated using a video-based walking track system. The gait parameters and the Achilles functional index (AFI) were further analyzed. The ankle joint angles of the injury side at mid-stance and pre-swing were highly correlated with the AFI. However, lack of sensitivity was found for the AFI. Increased measurement sensitivity of the Achilles tendon healing condition was found in the ankle joint angle of the involved side at the pre-swing and the level of asymmetry of the hindlimb joint position and stance/swing time. The overall sensitivity of the ankle motion analysis was significantly higher than that of AFI. We conclude that the ankle motion analysis is a reliable, reproducible, and sensitive tool for Achilles tendon analysis in rats.
This study investigates the interactions between cyclooxygenase (COX-2) and vascular smooth muscle cell (VSMC) phenotypic switching, the two important coupling mechanisms of the vasculature on arterial remodeling in response to high laminal shear stress. High aortic blood flow was induced by creating a fistula in the abdominal aorta and the adjacent IVC of anesthetized rats. Celecoxib, a selective COX-2 inhibitor (25 mg/kg/day), was fed in the chow, and animals were killed 8 weeks later. Blood flow, vasoreactivity and morphological changes in the aorta proximal to the fistula were measured. Concentrations of collagen, expression of desmin and smooth muscle myosin heavy chain (SM-MHC)-II in the aorta were determined. Celecoxib significantly increased aortic blood flow and reduced the contraction responses of aorta. Decreased medial thickness, presence of intimal thickening and derangement of elastic lamina were found in the aortic section of celecoxib-treated animals. Celecoxib significantly reduced the tissue content of collagen and upregulated expression of SM-MHC-II and desmin in the high-flow aorta. Inhibition of COX-2 enzymatic activity in the aorta exposed to higher blood flow resulted in increased blood flow and vascular remodeling. These functional changes were accomplished by VSMC phenotypic switching and reduced biosynthesis of collagen.
BackgroundIncreased incidence of intraoperative awareness was reported in critically ill patients during major operations, particularly under total intravenous (TIVA) or endotracheal general anesthesia (ETGA). However, the incidence and effect of anesthesia techniques on awareness in generally healthy, non-critically ill patients during operations have yet to receive significant attention.Methods and resultsThis retrospective matched case-control study was conducted between January 2009 to December 2014. Surgical patients (ASA physical status I-III) whom reported intraoperative awareness during this study period were interviewed and their medical records were reviewed. The potential risk factors for awareness were compared with the non-case matched controls, who were randomly selected from the database. A total of 61436 patients were included and 16 definite cases of intraoperative awareness were identified. Patients who received ETGA and TIVA had significantly higher incidence of developing awareness compared to those who were anesthetized using laryngeal masks (LMA) (P = 0.03). Compared with the matched controls (n = 80), longer anesthesia time was associated with increased incidence of awareness (odds ratio 2.04; 95% CI 1.30–3.20, per hour increase). Perioperative use of muscle relaxant was also associated with increased incidence of awareness, while significantly lower incidence of awareness was found in patients who were anesthetized with volatile anesthetics.ConclusionsThe overall incidence of awareness was 0.023% in the ASA≤ III surgical patients who received general anesthesia. Anesthesia with a laryngeal mask under spontaneous ventilation and supplemented with volatile anesthetics may be the preferred anesthesia technique in generally healthy patients in order to provide a lower risk of intraoperative awareness.
