Fluorescence anisotropy imaging is used to measure the changes in compaction of undamaged chromatin upon induction of clustered double-strand breaks. Live-cell dynamics is combined with subsequent immunofluorescence. PCNA is recruited immediately at the site of damage, while forming foci of repair away from the site at later time points in regions of open chromatin.
