ABSTRAKPenggunaan obat tradisional sangat sering dijumpai, karena penggunaannya yang bebas tanpa harus berkonsultasi dengan tenaga medis, sehingga masih didapati penggunaan bahan kimia obat dalam jamu. Bahan kimia obat yang sering ditambahkan dalam jamu adalah parasetamol dan fenilbutazon. Pada penelitian ini digunakan sistem KLT Densitometri untuk mendeteksi parasetamol dan fenilbutazon, agar mendapatkan sistem KLT yang optimal, validasi meliputi selektivitas, linearitas, akurasi, presisi, batas deteksi dan batas kuantitasi, dan penetapan kadar parasetamol fenilbutazon dalam sampel jamu pegal linu. Fase gerak yang optimal dalam deteksi parasetamol dan fenilbutazon yaitu etil asetat : kloroform (2:1). Dengan panjang gelombang maksimal parasetamol 240 nm, dan fenilbutazon 237 nm. Metode yang digunakan memiliki selektivitas, linearitas dan memenuhi kriteria akurasi dan presisi, tetapi pada batas deteksi dan batas kuantitasi belum memenuhi sesuai dengan yang dipersyaratkan. Pada 30 sampel jamu, ditemukan 5 sampel positif mengandung fenilbutazon yaitu j, k, s, u, v dengan persen kadar 9,5053%; 10,6138%; 62,8776%; 42,8839% dan 24,9238%. ABSTRACTTraditional medicine or commonly known as herbal medicine is often found, because its free to use without consulting medical therefore. Chemical drugs in the herbal products are still often found. Drug chemicals often added to herbs are paracetamol and phenylbutazone. In this study TLC Densitometry system used to detect paracetamol and phenylbutazone, to get optimal TLC system, validation includes selectivity, linearity, accuracy, precision, LOD and LOQ, determination of paracetamol and phenylbutazone in herbal samples.Optimal mobile phase for detection paracetamol and phenylbutazone is ethyl acetate : chloroform (2:1). Maximum wavelength of 240 nm paracetamol, 237 nm phenylbutazone. The method used has selectivity, linearity and met the criteria of accuracy and precision, but the limit detection and limit quantitation do not met requirements.
