A procedure for the simultaneous determination of secretion rates (S.R.) of deoxycorticosterone (DOC) and corticosterone (B) by double-isotope derivative assay is described. In a detailed assessment of the method the importance of careful purification and isolation of the metabolites of both steroids is emphasized. The S.R. of DOC was 174.6 ± 14.6 S.E. μg/24 h in 19 determinations in 15 normal controls and 7712 ± 2072 S.E. in three determinations after metapyron administration. Corticosterone S.R. was 2778 ≠ 232 S.E. μg/24 h in 15 determinations in 12 normal controls. All the experiments were done on normal sodium intake.
