We studied the phenotypic effects of polymorphisms at the MYF5 gene in a divergent F 2 swine population and found that one polymorphism was due to an insertion and another to a deletion. The genotypes of 359 F2 animals were obtained and the Normal/Normal (NN) and Normal/Insertion (NI) genotypes analyzed to determine associations with phenotypic data for performance, carcass and meat quality traits. Significant differences were observed (p < 0.05) between NN and NI animals for drip (NN = 3.14 ± 1.56; NI = 3.69 ± 2.78%), cooking (NN = 32.26 ± 2.41; NI = 33.21 ± 2.31%) and total loss (NN = 34.16 ± 2.63 and NI = 34.97 ± 2.08%). The Deletion marker was not statistically tested. The results indicate that the allelic variant Insertion is associated with a deleterious effect on meat quality traits and should be monitored in marker assisted selection programs.
The objective of this work was to evaluate 41 microsatellite markers for heterologous amplifications in piracanjuba (Brycon orbignyanus). Some markers were tested for the first time. Loci were optimized for PCR conditions and applied to a sample of 49 individuals. Thirty-one loci resulted in PCR product formation, whereas ten loci yielded intelligible polymorphic patterns in the evaluated sample and can be used for amplifications in this species. From the evaluated markers, four loci (BoM1, BoM13, Bh6, and Bh16) are valid to be applied in the study of piracanjuba.Index terms: Brycon orbignyanus, aquaculture, hatchery, molecular markers.
Otimização de microssatélites heterólogos em piracanjubaResumo -O objetivo deste trabalho foi avaliar 41 marcadores microssatélites para amplificação heteróloga em piracanjuba (Brycon orbignyanus). Alguns marcadores foram testados pela primeira vez. Os loci foram otimizados para condições de PCR e aplicados a uma amostra de 49 indivíduos. Trinta e um loci resultaram em formação de produtos de PCR, enquanto dez loci resultaram em padrões polimórficos inteligíveis na amostra avaliada e podem ser usados para amplificações nessa espécie. The first complete mitochondrial genome for the Anostomidae fish family has been announced. Piracema fishes are the potamodromous migratory species from South America, which undergo lengthy and dramatic yearly reproductive upstream runs. The piracema species Leporinus piavussu has recently been described after a long misidentification history. Its mitogenome, assembled using NGS data and Sanger sequencing, consists in a 16,682 bp circular molecule (GenBank accession number KM886569). The exact sequence and position of 37 mitochondrial genes and the control region is established. A possible case of heteroplasmy was found with NGS and corroborated by Sanger sequencing. These results will positively contribute to the debate about this group's taxonomy, evolution and conservation.
