Given the clinical importance of free thiamine and its esters on the essential human organs, their determination seems necessary to appreciate vitamin B1 status in physiological and pathological states of local populations. The present preliminary study describes a method of analysis of free thiamine and its phosphate esters: thiamine monophosphate and thiamine diphosphate by reverse phase liquid chromatography and fluorimetric detection. Validation tests were applied to verify the reliability of the proposed method. The results indicated that the chromatographic system was suitable for chromatographic analysis of free thiamine and its esters in whole. Thus, the method was applied to whole blood samples of 35 alcoholic patients under alcoholic detoxification cure in Abidjan. The results of the application showed that 100% of the alcoholic subjects presented a total thiamine concentration under the accepted norm of 20µg.L -1 . Before the cure at Day 0, 74,28% of the subjects presented a severe deficiency. After 30 days, all of the subjects still presented a marginal status of vitamin B1. Thiamine supplementation included in the alcoholic detoxification cure seems to improve its status. However, it seems that the latter should be of sufficient duration to ensure the return to an effective normal vitamin B1 status.
Les combinaisons thérapeutiques antipaludiques à base d'artéméther-luméfantrine sous forme de poudres lyophilisées, sont des préparations multidoses destinées aux enfants pour le traitement du paludisme. Dans le but d'obtenir une bonne observance thérapeutique chez les enfants, ces poudres lyophilisées à reconstituer ont été mises sur le marché. La stabilité des suspensions obtenues après reconstitution avec différents types d'eau accessible à toutes les couches sociales dans les zones endémiques de paludisme a été évaluée. Les caractères organoleptiques et le pH des suspensions ont été recherchés. Les teneurs en artéméther et en luméfantrine ont été recherchés respectivement par chromatographie liquide et par spectrométrie UV. Les produits de dégradation de l'artéméther et de la luméfantrine ont aussi été recherchés par chromatographie sur couche mince de gel de silice. Des études microbiologiques ont été réalisées pour la recherche des germes anaérobies viables totaux et la recherche d'entérobactéries. L'étude a révélé que les caractères organoleptiques et le pH de chaque suspension ont été acceptables. Les principes actifs n'ont pas été dégradés après reconstitutions des poudres lyophilisées dans tous les échantillons durant la période d'étude. Les concentrations obtenues en artéméther et en luméfantrine de tous les échantillons analysés, étaient conformes aux normes. Les échantillons n'ont pas été contaminés par des bactéries, des levures et des moisissures au cours de cette période d'étude.
Mareya micrantha (Euphorbiaceae) is an Ivorian medicinal plant widely used in gastrointestinal disorders. So as to understand and identify metabolites possibly implicated in the activity of the plant on intestinal muscles, this study has been undertaken. From the crude ethanolic extract EE of the leaves, hexanic (EH), chloromethylene (EC), ethyl acetatic (EA) and butanolic (EB) sub-extracts were prepared as well as the hydroethanolic residual phase dried (Ar) and their effects on the isolated duodenum of rabbits were determined. If the sub-extracts EH, EA and EB, but also Ar, confirmed the myostimulant activity of the crude extract EE, this was not the case of EC which showed myorelaxant activity. This sub-extract, subjected to purification, led to the isolation of three major triterpenoids type 29norcucurbitacin (1-3) whose structures were confirmed using the combined analyses of their spectroscopic data (UV, MS, 1D NMR and 2D). All these molecules have shown myorelaxant activity, the most significant one being 2 with an IC50 value of 36.9 µM. These molecules would act by blocking both intracellular and extracellular calcium.
