BackgroundNano-scale dendrimers are synthetic macromolecules that frequently used in medical and health field. Traditional anibiotics are induce bacterial resistence so there is an urgent need for novel antibacterial drug invention. In the present study seventh generation poly (amidoamine) (PAMAM-G7) dendrimer was synthesized and its antibacterial activities were evaluated against representative Gram- negative and Gram-positive bacteria.MethodsPAMAM-G7 was synthesized with divergent growth method. The structural and surface of PAMAM-G7 were investigated by transmission electron microscopy, scanning electron microscope and fourier transform infrared. Pseudomonas. aeruginosa (n = 15), E. coli (n = 15), Acinetobacter baumanni (n = 15), Shigella dysenteriae (n = 15), Klebsiella pneumoniae (n = 10), Proteus mirabilis (n = 15), Staphylococcus aureus (n = 15) and Bacillus subtilis (n = 10) have been used for antibacterial activity assay. Additionally, representative standard strains for each bacterium were included. Minimum Inhibitory Concentration (MIC) was determined using microdilution method. Subsequently, Minimum Bactericidal Concentration (MBC) was determined by sub-culturing each of the no growth wells onto Mueller Hinton agar medium. The cytotoxicity of PAMAM-G7 dendrimer were evaluated in HCT116 and NIH 3 T3 cells by MTT assay.ResultsThe average size of each particle was approximately 20 nm. PAMAM-G7 was potentially to inhibit both Gram positive and gram negative growth. The MIC50 and MIC90 values were determined to be 2–4 μg/ml and 4–8 μg/ml, respectively. The MBC50 and MBC90 values were found to be 64–256 μg/ml and 128–256 μg/ml, respectively. The cytotoxity effect of dendrimer on HCT116 and NIH 3 T3 cells is dependent upon exposure time to and concentration of dendrimers. The most reduction (44.63 and 43%) in cell viability for HCT116 and NIH 3 T3 cells was observed at the highest concentration, 0.85 μM after 72 h treatmentm, respectively.ConclusionsThis study we conclude that PAMAM-G7 dendrimer could be a potential candidate as a novel antibacterial agent.
Background: Nanoscale poly (amidoamine) dendrimers have been investigated for their biological demands, but their antibacterial activity has not been widely discovered. Thus, the sixth generation of poly (amidoamine) dendrimer (PAMAM-G6) was synthesized and its antibacterial activities were evaluated on Gram-negative bacteria; P. aeruginosa, E. coli, A. baumannii, S. typhimurium, S. dysenteriae, K. pneumoniae, P. mirabilis, and Gram-positive bacteria, and S.aureus and B. subtilis, which were isolated from different clinical specimens and standard strains of these bacteria. Methods: In this study, 980 specimens including urine (47%), blood (27%), sputum (13%), wounds (8%), and burns (5%) were collected from clinical specimens of 16 hospitals and clinics in city of Sabzevar, Iran. Then, the target bacteria were isolated and identified using standard methods. Minimum inhibitory concentration and minimum bactericidal concentrations against Gram-positive and Gram-negative bacteria were determined according to guidelines described by clinical and laboratory standards institute (CLSI). Standard discs were prepared using 0.025, 0.25, 2.5, and 25 μg/mL concentrations of PAMAM-G6 on Mueller-Hinton agar plates to determinate the zone of inhibition. The cytotoxicity of PAMAM-G6 dendrimer was evaluated in HCT116 cells by MTT assay. Results: The most important isolated bacteria were E. coli (23.65%), S. aureus (24.7%), P. aeruginosa (10.49%), B. subtilis (7.7%), S. typhimurium (8.87%), A. baumannii (7.02%), K. pneumoniae (7.1%), P. mirabilis (6.46%), and S. dysenteriae (3.6%). Moreover, it was found that poly (amidoamine)–G6 exhibited more antibacterial efficacy on standard strains than isolated bacteria from clinical samples (p<0.05). The cytotoxicity of PAMAM-G6 to the cells showed that cytotoxicity depended on the concentration level and exposure time. Conclusion: The PAMAM-G6 dendrimer showed a positive impact on the removal of dominant bacterial isolated from clinical specimens and standard strains.
Background: The aim of this study was to evaluate the physical and chemical quality of groundwater in the villages of Qom province during summer 2014, and to compare the results with Iran's Standard No. 1053. Methods: This research is a descriptive sectional study. During the investigation, one step sampling with the necessary repetitions (if necessary) was performed of each well for evaluation of physical and chemical quality of groundwater in the villages of Qom province. SPSS version 16 and ARCGIS software were employed for data analysis and data zoning respectively. Results: The average concentration of magnesium, sulfate and ammonia in wells water were higher than standard. The average concentration of fluoride was lower than the standard limit. pH and turbidity in all wells were within the standard limit but sulfate in 33.33%, ammonia in 13.33%, magnesium in 10.66%, sodium in 40%, calcium in 5.33%, nitrate in 17.33%, total dissolved solids (TDS) in 14.66% and electrical conductivity (EC) in 24% of wells were higher than the standard limit, and fluorine in 98.66% of wells was lower than the standard limit. The results of zoning in the villages of Qom province revealed that measured values at most wells were in the maximum amount in central and northeastern areas and minimum amount in southern and western areas. Conclusion: This study indicated that most of the physicochemical parameters such as fluoride in almost all villages, and sulfates, chloride, magnesium, sodium, calcium, nitrate, TDS and EC in some of the wells did not fall within the permissible limit.
Background and Purpose: Improper wastewater and solid waste management in rural areas could be a risk to human health and environment pollution. One percent of Iran's rural area is connected to the wastewater collection network. Solid waste management in rural areas of Iran is mainly consisted uncontrolled dumping and open burning. The aim of this study is prioritization of wastewater and solid waste separate and combined management strategies in rural areas of Iran. Materials and Methods: This was a descriptive study. In this study, firstly were determined appropriate and conventional methods for wastewater and solid waste separate and combined management by using national and case studies. Then, using specified criteria and by applying a weighting system, prioritization was conducted and implementation strategies presented for wastewater and solid waste separate and combined management. Results: The first priority for the collection and treatment, wastewater in rural areas are smalldiameter gravity systems and preliminary treatment with complementary treatment by land, respectively. In order to the rural solid waste management, organic compost complementary systems were in first priority. In the wastewater and solid waste combined management, the first priority was compost and biogas production by combining anaerobic UASB reactor and Chinese biogas. Conclusion: Considering for influence of various factors in selecting an appropriate method is very important in order to wastewater and solid waste separate and the combined management of a rural. Therefore, the accordance of presenting strategy with local conditions and facilities should be taken into consideration.
