Local cerebral metabolic rates for glucose were studied by [14C]-2-deoxyglucose autoradiography in adult rats following acute and chronic unilateral deafferentation, with particular attention to the barrel field regions of the primary somatosensory cortex. Deafferentation was produced by permanently removing all of the large whiskers (vibrissae) on one side of the face. Data from experimental animals were then compared to data from sham-operated controls at 1, 5, 10, 15, 30, and 60 days after deafferentation. The rate of glucose utilization was maximally depressed at day 1 in the deafferented barrel field. After that, there was a progressive recovery of glucose utilization toward control levels at each subsequent time point. In contrast, glucose utilization in the barrel field associated with the intact set of whiskers increased by day 5 and remained elevated throughout the duration of the experiment. Similar patterns of altered cerebral metabolism were observed following unilateral infraorbital nerve transection. These results demonstrate that interference with normal somatosensory input causes a transient decrease in glucose metabolism of the contralateral cortical barrel-field and, in addition, causes long-term increments in glucose metabolism in the ipsilateral cortical barrel field--a structure not normally influenced by acute manipulation.
Thirty min prior to anaesthetic induction for surgery, children aged 4-12 years old were given a 10 micrograms.kg-1 oral transmucosal fentanyl citrate (OTFC) and were instructed to suck the OTFC until pruritus appeared (Group 2) or until the entire dose was consumed (Group 1). Sedation, apprehension and cooperation scores were rated, and vital signs including oxygen saturation were monitored until anaesthetic induction. The results showed that pruritus was present in 76% of children; however; in all but one child, it occurred after the OTFC had been completely consumed. There were no significant changes in oxygen saturation, but respiratory rate decreased from 19.6 +/- 1.7 to 18.4 +/- 1.3. Activity decreased significantly; however, cooperation and apprehension did not change. The conclusion was that pruritus cannot be used as an endpoint for OTFC effectiveness; however, OTFC dosed at 10 micrograms.kg-1 is effective in providing sedation without causing clinically significant changes in vital signs or oxygen saturation.
SimYard is a stochastic shipyard simulation tool designed to evaluate the labor costs of executing different schedules in a shipyard production environment. SimYard simulates common production problems such as task delays and labor shortages. A simulated floor manager reacts to problems as they arise. Repeatedly simulating multiple schedules allows the user to compare the schedules on many different metrics, such as expected labor costs and the probability of missing the deadline. A SimYard simulation is driven by many inputs that describe the shipyard being simulated. Determining the correct values for these inputs can be framed as a multivariate calibration problem, which can be solved using inverse regression methods. Predictive sampling from the resulting model provides an appropriate adjustment for statistical uncertainty.
