The purpose of this study was to examine the efficacy of a herbal combination of avocado, mung bean sprouts, and holy basil (Jamu ATOKE) in optimizing the reproductive health of adult female rats. Eighteen female Sprague Dawley rats aged 9-10 weeks old weighing around 180-250 g, and had not been pregnant previously were equally and randomly divided into 3 groups: one control group and two treatment groups (n= 6 rats per group). The treatment groups received Jamu ATOKE with a concentration of 2.5 and 5%, respectively. Jamu ATOKE was added to the drinking water and consumed by the rats for 30 days prior to pregnancy. Reproductivity (pregnancy success, estrogen and progesterone hormone concentrations), and health performance (body weight gain, food, and water consumption, motor activity, red blood cell (RBC) count, white blood cell (WBC) count and differentials, SGPT, SGOT, ureum, and creatinine) of rats were observed. The results showed that groups receiving the Jamu ATOKE had better reproductive and health performance compared to those of the control group. The administration of Jamu ATOKE can significantly increase the concentration of estrogen and progesterone in rats (P0.05). It can also improve the fertility and health of the female rats prior to pregnancy.
Penelitian ini dilakukan untuk mengeksplorasi khasiat fitofarmaka lokal Indonesia yaitu kemangi sebagai antifertilitas dalam meningkatkan kesehatan dan keamanan tikus betina. Sebanyak 15 ekor tikus betina galur Sprague-Dawley (umur 8 minggu, bobot badan 220-230 g) dibagi acak ke dalam 3 perlakuan dan 5 ulangan, yaitu: kontrol (K0); kemangi 1% (K1); dan kemangi 5% (K5) dalam bentuk maserasi yang ditambahkan ke dalam air minum. Pemberian jamu dilakukan selama 20 hari dimulai pada fase diestrus. Parameter profil darah yang diamati adalah gambaran darah merah (jumlah butir darah merah, hematokrit, dan hemoglobin) dan diferensial darah putih (limfosit, monosit, neutrofil, eosinofil, basofil). Parameter keamanan diamati dengan melihat fungsi organ hati (SGPT, SGOT) dan ginjal (ureum, kreatinin). Hasil menunjukkan bahwa profil darah dan pengamatan fungsi hati dan ginjal pada kelompok tikus betina yang diberi kemangi tidak terdapat perbedaan nyata (P>0.05) dengan kelompok kontrol. Pemberian kemangi tidak memengaruhi kondisi fisiologis tikus betina dan tidak toksik sehingga dapat digunakan untuk meningkatkan kesehatan tikus betina.
Mortality and health disturbances in children often correlate with maternal health and fertility. Avocado, mung bean sprouts, and holy basil have been traditionally used to improve maternal health, before and during pregnancy. Eighteen female Sprague Dawley rats (9–10 weeks old, BW: 180–250 g) were randomly divided into 3 treatment groups (n=6 rats for each group) of control, group I and II. ATOKE were added into rats drinking water and consumed for 30 days before pregnancy. After the rats were pregnant and gave birth, pup per parent ratio, pup mortality, and pup health performance (body weight gain, feed and drink consumption, motor activity, pup speed in finding light and feed, red blood cell (RBC) count, white blood cell (WBC) count and differential, SGPT, SGOT, blood urea nitrogen (BUN), creatinine, and proinflammatory mediators (TNF-α and IL-6) were analyzed. Pups born to in group I had a generally better health performance compared to the control group. There was a 100% mortality in the litter of group II due to the excessive aggressive behavior of the dams as a sign of toxicity. Administration of 2.5% Jamu ATOKE showed the best effects on the health and intelligence of the rat pup.
