Introduction: Cataracts caused by old age are one of the most frequent causes for blindness and poor vision worldwide. In this study, we aimed to clarify the possible role of rs1894720 polymorphism in the pathogenesis of age-related cataract. Material and methods: Rs1894720 polymorphism genotype was detected by TaqMan. Bioinformatics analysis, luciferase assay, real-time PCR, western blot, and protein density analysis were conducted to establish the correlations between MIAT and miR-26b as well as between BCL2L2 and miR-26b. Flow cytometry and MTT assay were also performed to observe the effect of MIAT/miR-26b/BCL2L2 signalling pathway on the status of cell apoptosis and viability. Results: MIAT functioned as an endogenous competing RNA to sponge miR-26b. In addition, BCL2L2 was identified as a target of miR-26b. Therefore, the expression of miR-26b was obviously suppressed by MIAT or anti-miR-26b, while the mRNA and protein expression of BCL2L2 was up-regulated in the presence of MIAT or anti-miR-26b. Moreover, the positive effect of MIAT on BCL2L2 expression was exerted via inhibition of the expression of miR-26b. In addition, the cells transfected with MIAT or anti-miR-26b showed suppressed expression of caspase-3 and reduced apoptosis index but higher cell viability, indicating that MIAT could suppress cell apoptosis via inhibition of miR-26b expression. Furthermore, the subjects carrying the GT and TT genotypes of single-nucleotide polymorphism (SNP) rs1894720 were associated with a higher risk of age-related cataracts, as indicated by their odds ratio (OR) and p values. Conclusions: Rs1894720 SNP could down-regulate the expression of MIAT, thus leading to reduced BCL2L2 expression and enhanced epithelial cell apoptosis in the lens, eventually increasing the incidence of age-related cata ract.
IntroductionThe signaling pathway of IL-6 has been reported to be associated with the severity of glaucoma. And the rs12976445 SNP in miR-125 could alter the expression of miR-125, which is directly targeted by IL-6R.Material and methodsIn this study, we recruited 88 POAG patients and grouped them according to their genotype of rs12976445 as GG group, GC group and CC group to study the association between the miR-125a polymorphism and POAG. We collected demographic characteristics and peripheral blood samples from 88 subjects. Then, rs12976445 genotypes in these subjects were determined to evaluate their relationship with the POAG index. THP-1 and U937 cells were transfected with miR-125a mimic or IL-6R siRNA to assess the relationship between miR-125a and IL-6R/ACHE.ResultsIL-6R is a downstream target of miR-125a and the overexpression of miR-125a showed significantly decreased mRNA and protein levels of IL-6R. The expression levels of miR-125a were the highest in the GG group and the lowest in the CC group, while the activity of IL-6 was comparable in the three groups. Moreover, the mRNA and protein levels of IL-6R were the lowest in the GG group and the highest in the CC group. Additionally, significantly thinner RNFL, larger average cup disc ratio, larger vertical cup disc ratio, and depressed visual field were observed in POAG patients carrying the CC genotype.ConclusionsIn summary, our data suggested that the rs12976445 polymorphism was significantly associated with the risk of POAG.
Introduction To observe the short-term and long-term effects of extensive fascial resection combined with autologous limbal stem cell sharing transplantation for pterygium. Methods A retrospective case series analysis was performed with 403 patients (420 eyes) who underwent extensive fascial resection combined with autologous limbal stem cell sharing transplantation in the Department of Ophthalmology, Second People's Hospital of Yunnan Province from January 2017 to June 2018. Before, and 10 days, 1 month, 3 months, and 6 months after surgery, the tear film rupture time (BUT), the height of the tear river (SLT), and the anterior segment of the fluoroscopy staining were recorded to record the scar on the conjunctiva and corneal surface. Patient self-consciousness and satisfaction with the appearance of the survey. Results BUT after surgery was prolonged than before operation, and there was no significant difference in tear height preoperative and postoperative. The postoperative follow-up period (6 months), the recurrence rate of pterygium was 0.21%.Patients have higher satisfaction with appearance, and have no obvious foreign body sensation, photophobia, tearing and other subjective symptoms. Discussion and conclusion Extensive fascial resection combined with autologous limbal stem cell sharing transplantation for the treatment of pterygium, the patient's appearance satisfaction is higher, and the recurrence probability is lower than the average recurrence rate of limbal stem cell transplantation. The operation is simple and easy to learn. This is a surgical procedure worth promoting.
