This research aims to looking for 2 candidates from the new probioticlactic acid bacteria fermented food origin. This study uses seven isolates were isolated from whey, cassava, and fish budu. 7 isolates have been tested potential as a producer of glutamic acid but not known as a candidate probiotic petensinya. This study uses a completely randomized design, each repeated 3 times with 5 stages of the research are: 1) Testing capability isolates at low pH (pH 2.5); 2) testing the ability of isolates in bile (0.3 and 0.5%); 3) Viability isolates 4) Testing capability isolates in killing the pathogenic bacteria (Escherichia coli, Staphylococcus aureus and Salmonella enteritidis). The results showed the 7 isolates can be used as a probiotic candidates with the results of the resistance to pH 2.5 for 3 hours is 63.91-92.75% and the 6 hour incubation is 48,76-72.44%, resistance to bile salt concentration of 0.3% showed resistance to 22.86-83.57% and a concentration of 0.5% showed resistance 18.35-72,77%, 81.05-92.67% viability and inhibitory effect on the bacteria Escherichia coli is 7:33 to 12:01 mm, is 11:11 to 14:05 mm Staphylococcus aureus and Salmonella enteritidis is 12:02 to 18:08 mm. From 7 isolates after penseleksian ability as a candidate probiotic mixture then isolate F6 from durian fermented and isolate C8 from buffalo milk fermented (Dadih) are able to live at pH 2.5 for 3 hours of 92.75% and 86.06%, at 6 hours of 72.44 and 71.42%, the bile salt 0.3% were 83.57% and 78.75% and the concentration 0.5% are 77.17% and 72.77%, with viability were 92.67% and 92.23%, while the ability to kill pathogens such as Escherichia coli was 10.49 and 8.89 mm, Staphylococcus aureus was 14.05 and 13.70 mm, dan Salmonella enteritidis was 18.08 and 14.18 mm.
An experiment has been conducted to study the effect of diets with different level of protein on the production performance of offspring (F1) crossbred between arab and kampung chicken. Fifty four crossbreed hens of 20 weeks of age were used in the experiment. The hens were divided into 3 feeding groups, each 6 x 3 heads. Treatment diets were formulated in three different levels of protein: 15, 17 and 19 %. The diets which an average energy content of 2850 kcal/kg were offered for 8 weeks. Variable observed included: feed intake, hen day egg production, FCR, egg weight and color index of yellow egg. The data were statistically analyzed by variance analysis in completely block design with 3 treatments and 6 replicates. The results indicated the protein level had significant effect on all variables. Diet with 19 % crude protein gave the best performance.
An experiment was conducted to study the ffict of feeding by-product from virgin coconut oil processing (blondo) in ration on total colony oflactobacillus sp, Salmonella sp and Escherichia coli in ileum of broiler. Broilers reared for 6 weel
This research aimed to identify polymorphism of the GH gene in Sikumbang Jonti ducks using the PCR-RFLP (polymerase chain reaction-restriction fragment length polymorphism) method as a candidate for genetic marker selection. This research used 123 blood samples of male and female Sikumbang Jonti ducks. The ducks were eight weeks old and kept intensively at the UPT Fakultas Peternakan Universitas Andalas. Blood samples of Sikumbang Jonti ducks were taken through the brachial vein as much as ± 2 mL. The Genomic DNA Purification Kit (Promega) protocol isolated DNA from blood samples. A pair of primers F amplified the total DNA: 5'-GGA CAG CCT GAG GAA AGA GT-3 'and R: 5'-GTG GAA GGT GGG GAG ACT TC-3', which produced a fragment region of the GH gene exon 3 with a cut region of part intron two and part intron 3 with a primary length of 833 bp. The amplified product was restricted by the enzyme TasI which recognizes the cutting site (↓AATT). Based on the restriction results, two (2) genotypes were obtained, namely homozygous (-/-) as many as four (4) and homozygous (+/+) as many as 119. The analysis of restriction products included the allele frequency, namely the allele (+) of 0.98 and the allele (-) of 0.02, and genotypes frequency of homozygous (-/-) of 0.03 and homozygous genotypes (+/+) of 0.97. The diversity of the GH gene is in the Hardy-Weinberg equilibrium. Based on the research results, it can be concluded that there is diversity in the GH gene of the Sikumbang Jonti duck, and it is in the Hardy-Weinberg equilibrium, so it can be used as a candidate for the genetic marker of ducks.
