Hybrid genes were constructed to express bifunctional hybrid proteins in which staphyloccal nuclease A with or without an amino-terminal OmpA signal sequence was fused with TEM beta-lactamase (at the carboxyl terminal side) using the signal peptide of the major outer membrane lipoprotein of Escherichia coli as an internal linker. The hybrid proteins were found to be inserted in the membrane. Orientation of the hybrid protein with the OmpA signal peptide showed that the nuclease was translocated into the periplasm and the beta-lactamase remained in the cytoplasm. This indicates that the cleavable OmpA signal peptide served as a secretory signal for nuclease and the internal lipoprotein signal served as the transmembrane anchor. In the absence of the OmpA signal sequence the topology of the hybrid protein was reversed indicating that the internal lipoprotein signal peptide initially served as the signal peptide for the secretion of the carboxy terminal beta-lactamase domain across the membrane and subsequently as a membrane anchoring signal. The role of charged amino acids in the translocation and transmembrane orientation of membrane proteins was also analysed by introducing charged amino acids to either or both sides of the internal lipoprotein signal sequence in the bifunctional hybrid proteins in the absence of the amino-terminal signal sequence. Introduction of two lysine residues at the carboxy-terminal side of the internal signal sequence reversed the topology of the transmembrane protein by translocating the amino-terminal nuclease domain across the membrane, leaving the carboxyl terminal beta-lactamase domain in the cytoplasm. When three more lysine residues were added to the amino-terminal side of the internal signal sequence of the same construct the membrane topology flipped back to the original orientation. A similar reversion of the topology could be obtained by introducing negatively charged residues at the amino-terminal side of the internal signal sequence. Present results demonstrate for the first time that a bifunctional transmembrane protein can be engineered to assume either of the two opposite orientations and that charge balance around the transmembrane domain is a major factor in controlling the topology of a transmembrane protein.
Given the increasingly serious nature of environmental problems, many countries have recently declared carbon neutrality policies and expended efforts to implement them. The domestic building industry aims to reduce its environmental impact using life-cycle assessments (LCAs) of buildings according to the Green Standard for Energy and Environmental Design. However, it is difficult to perform efficient LCAs because the required quantity takeoff process is complex, and the quantity takeoff sheet may not exist during the building’s design phase. In this study, 21 building LCAs were used to simplify and improve the efficiency of the proposed method and enable building LCAs even when there was no quantity takeoff sheet. Furthermore, a standard quantity database of building materials was constructed based on the analysis of the input quantities of building materials per unit area, and the apartment buildings LCA method was proposed using this database. The input quantities of building materials were analyzed using the probabilistic analysis technique. The probability distribution was derived using Monte Carlo simulations, and the goodness-of-fit was verified. Finally, the reliability of the proposed building LCA method was verified using a case study.
The purpose of study was to introduce menus consisting of traditional local foods consumed in Busan, Korea. Five different types of formal meal menus with three side dishes and five different types of informal meal menus of which the main dish was not cooked rice were developed. Scores for the menu evaluations of formal and informal meals were higher than 7.9 (mean score of 6 category) on a 9.0 scale, suggesting that the menus were well constituted in terms of 'repeated use of ingredients and cooking methods', 'balance between main and side dishes', 'frequency of usage of local products', 'nutritional balance', 'variety of dishes', and 'popularization'. Nutritional values of menus were compared with those of the KDRI for men aged 30~49 years old. All ten menus reflected a low calorie, high protein, and high fiber diet. Mineral contents (Ca, Fe, Zn, K, and Na) of the above 10 menus were higher than those of the KDRI. On the other hand, vitamin contents slightly differed according to meal type due to limited fresh vegetable usage in the winter season. Vitamin A, B (B1, B2, niacin, B6), C, folic acid, and E contents in the five informal menus were found to be proper or greater than those of the KDRI. However, for the formal meal menus, all vitamin contents except that of vitamin A in menu typeⅠ was lower than that of the KDRI. Content of vitamin C in menu type V was approximately 50% of that of the KDRI. The most preferred formal menu was consisted of Ogokbap, Boreumjijimi, Dongchimi, Namul (nine varieties), Gaksaeksanjeok (Shark/Daegu/Gunso), and Gimgui. And that for informal menu was consisted of Honghapjuk, Baechu kimchi, Dongchimi, Miyeokseolchi, Kodari jorim, and Neulgeun hobakjeon. Suggestions for providing more fresh vegetables or fruits for certain types of meals (formal meal type Ⅴ) were offered. Additionally, pre-preparation of sea products were required to reduce Na contents.
Shotcrete should be attached to the ground and should have stable strength for a long-term. It should develop strength earlier for rapid work. Therefore, in this study, three types of accelerators—aluminate, cement mineral, and alkali-free—were selected and mixed to secure the initial strength. Depending on the type and mixing rate of each accelerator, slump, air amount, and compressive strength were used to evaluate the basic properties, boiling water absorption test, and chloride ion penetration resistance to conduct durability analysis. The mixing of aluminate-based and cement-mineral-based accelerators was effective in improving the initial strength, and alkali-free accelerator was effective in improving the long-term strength. The mixture to which accelerators were not mixed showed the highest water-tightness.
The increase in demand for leisure time by the public and the vitalization of agricultural tourism resulting from the expansion of the five-day work week are expected to greatly contribute to the rural economy, but actual studies analyzing the ripple effect of the agricultural tourism industry are lacking. In order to analyze the economic ripple effect, tourism or agricultural tourism must become an independent sector in the inter-industry relations table's sector classification, but because it is not, the study uses a method of analysis which involves creating a satellite account. However, because agricultural tourism has a clear main agent in farms unlike general tourism which does not, there is one method in which provisions are made by farms or farm villages and another method in which outside products are used. The purpose of the present is to measure the economic ripple effect of agricultural tourism with a focus on 162 subclasses by applying positive data from the education sector input into agricultural tourism. Satellite accounts which considered intermediate input were created and applied to positive data, the analysis of which revealed agricultural tourism to account for 462 billion won, which is 0.01% of the total production amount of 3,503,480 billion won, while the production inducement coefficient was 3.2895 units when the final demand of agricultural tourism occurs. When the production inducement coefficient is at 3.2895 units, highest sector was agricultural tourism at 0.9968 units followed by restaurants and bars at 0.3325 units, roadt transportation services at 0.3183 units, lodging services at 0.1520 units, and petroleum products at 0.1290 units.
