Objectives This study aims to evaluate the antioxidant and antiviral potency of n-hexane, ethyl acetate and, water fractions of Begonia medicinalis Ardi & D.C.Thomas as well as to identify the chemical constituents. Methods Assays for antioxidant and antiviral activity (HIV-1) were carried out on MT-4 cells infected with HIV using the DPPH method and the determination of the cytopathic effect. Meanwhile, GC-MS was used to identify the chemical compounds. Results The determination of antioxidants showed that all fractions possessed potent activity with the IC50 ranging from 2.61 to 8.26 μg/mL. From the antiviral activity of MT-4 cells infected by HIV, the n-hexane fraction of B. medicinalis showed the most potency with the IC50 of 0.04 ± 0.05 μg/mL. It has less cytotoxicity (11.08 ± 4.60 μg/mL) affording the high selectivity index of 238.80. Furthermore, GC-MS analysis of n-hexane fraction found the major compound of carboxylic acid derivate with the area percentage of 76.4% and the presence of phenolic compounds (8.38%). Meanwhile, in water fraction, terpenoids were found in a higher concentration (10.05%) than others. Conclusions Therefore, this study supports the application of B. medicinalis as a herbal medicine for antioxidant and antiviral.
Hyperglycemia is a condition of increased blood glucose levels caused by insufficient insulin enzymes produced to control blood sugar levels in the body. This study aimed to determine the activity of ethanol-water, nhexane, and ethyl acetate extracts of Dioscorea alata L. in reducing blood glucose levels in hyperglycemia modeled mice that were given intraperitoneal alloxan induction. 25 white rats were divided into 5 groups of test animals. Each group consists of 5 mice. The first group was given 0.5% sodium carboxymethylcellulose solution as a placebo, the second group was given 0.65 mg/kg BW glibenclamide as a positive comparison group, the third group was given the suspension of ethanol-water extract, the fourth group was given n-hexane extract, and the fifth group was given ethyl acetate extract at a dose of 140 mg/kg BW. The test preparation was orally given within 14 days of testing. The test was measured as the initial blood glucose levels of mice after alloxan induction and after giving the test preparation suspension. The results of the qualitative analysis illustrate that the water-ethanol extract can reduce blood glucose levels by 31.39%, n-hexane extract by 51.11%, and ethyl acetate extract by 50.77%. The results of quantitative analysis using One Way ANOVA method showed that the water-ethanol, n-hexane, and ethyl acetate extracts significantly decreased the blood glucose levels of mice. The best antidiabetic activity was ethyl acetate extract with a decrease in rat blood glucose levels by 169 mg/kg BW.
Luka diabetik merupakan luka yang disebabkan oleh infeksi akibat dari tingginya glukosa darah ? 200 mg/dL. Daun Diospyros celebica Bakh., memiliki potensi antibakteri, antidiabetes, antivirus, dan antijamur. Tujuan penelitian ini adalah menentukan aktivitas penyembuhan luka diabetik pada tikus putih yang di induksi aloksan dengan diberikan terapi ekstrak daun eboni. Induksi diabetes dilakukan menggunakan aloksan dosis 140 mg/kgBB intraperitonial. Tikus dilukai pada bagian punggung dengan diameter luka ± 1 cm dan kedalaman 2 mm. Jumlah sampel dalam penelitin ini adalah 25 ekor tikus dibagi menjadi 5 kelompok uji yaitu kelompok 1 diberikan suspensi glibenklamid secara oral dan gel plasebo pada luka, kelompok 2,3 dan 4, kelompok yang diberikan suspensi glibenklamid secara oral dan gel ekstrak daun eboni konsentrasi 5 %, 10 % dan 15 % pada luka), kelompok 5 (pemberian suspensi Na-CMC 0,5 % secara oral dan gel plasebo pada luka). Pengujian dilakukan selama sepuluh hari dengan dosis dua kali pemberian setiap hari. Pengamatan makroskopik, panjang luka diukur dan dilaporkan sebagai persentase penyembuhan luka. Hasil penelitian menunjukkan bahwa gel ekstrak daun eboni memiliki kemampuan untuk memperbaiki lesi pada luka diabetes pada tikus putih, konsentrasi gel ekstrak daun eboni yang paling efektif untuk memerbaiki lesi luka diabetik adalah konsentrasi 10%, dengan persentase penyembuhan 94,34%.
