Orchids of the Vanda helvola genus are extensively grown as ornamental plants. The method of propagation of plant seeds in tissue culture is an alternative that may be employed in the provision of a short period. Murashige and Skoog (MS) media and activated charcoal can optimize orchid cultivation utilizing tissue culture techniques. One of the keys to success in tissue culture is the application of natural types of growing regulatory substances (ZPT) at the proper concentration, which can lower the cost of orchid tissue culture.The objective of this study was to see how a combination of Murashige and Skoog (MS) media and activated charcoal influenced the Vanda helvola orchid subculture in vitro. The researchers employed a Complete Randomized Design (RAL) with four treatments and three repeats: negative control (K), ¼ MS (K1), ½ MS (K2), and 100% MS (K3) (K3). The observed parameters include the percentage of live explant, number of roots, and height of orchid plants. The results showed that the explant had a 100% life percentage of over 21 HST and had no noticeable influence on the number of roots and the plant's height.
Bacillus megaterium merupakan jenis bakteri endofit yang mempunyai potensi untuk dikembangkan sebagai bahan baku pupuk hayati. Selain mampu memaksimalkan penyerapan nutrisi tanaman, B. megaterium juga dapat menghambat pertumbuhan patogen tanaman, termasuk pada tanaman cabai. Tanaman cabai (Capsicum frutescens) masih dianggap sebagai komoditas yang menguntungkan, di samping karena permintaannya yang terus meningkat setiap tahun. Oleh karena itu, penelitian ini bertujuan untuk menguji aktivitas antagonis B. megaterium terhadap Fusarium oxysporum dan mengamati pengaruhnya pada pertumbuhan tanaman cabai rawit. Metode yang digunakan yaitu uji antagonis dengan teknik dual culture, dan pengaplikasian B. megaterium ke tanaman cabai rawit dengan variasi dosis, meliputi K, A1, A2, A3 dan A4. Berdasarkan penelitian, B. megaterium mampu menunjukkan penghambatan terhadap F. oxysprorum sebesar 67% pada hari ke sepuluh. Selain itu, tanaman cabai rawit yang diaplikasikan B. megaterium memberikan pertumbuhan yang baik dibandingkan kontrol untuk variabel tinggi tanaman, jumlah daun, Panjang akar, berat segar dan berat kering tanaman. Dari variasi dosis yang diberikan, perlakuan A4 dengan dosis 25 mL mampu memberikan pertumbuhan terbaik. Hal ini menunjukkan bahwa B. megaterium bersifat antagonis terhadap F. oxysporum dan mampu meningkatkan pertumbuhan tanaman cabai rawit.
Jamu gendong termasuk obat tradisional yang umum dikonsumsi masyarakat Indonesia, salah satunya jamu kunir asem. Tujuan penelitian ini mendeteksi ada tidaknya bakteri Escherichia coli dan Salmonella pada jamu kunir asem di Daerah Gamping. Metode yang digunakan adalah MPN (Most Probable Number), menghitung total Coliform pada jamu kunir asem. Uji positif Coliform selanjutnya di deteksi Escherichia coli dan Salmonella pada medium Endo agar dan Salmonella shigella agar, berturut-turut. Ke 6 sampel dari total sembilan sampel jamu kunir asem terkontaminasi bakteri Coliform dengan jumlah antara 7,40x10 hingga 1,10x104 Sel/mL. Deteksi Escherichia coli dan Salmonella pada sampel kunir asem memberikan hasil negatif.
Tanaman ciplukan memiliki senyawa yang berpotensi sebagai antibakteri. Potensi senyawa antibakteri kemungkinan juga dimiliki oleh bakteri endofit tanaman ciplukan. Penelitian ini bertujuan untuk mengisolasi bakteri endofit dari daun ciplukan yang berpotensi sebagai antibakteri terhadap Escherichia coli dan Staphylococcus aureus. Metode penelitian meliputi sterilisasi, pembuatan media, pengambilan sampel, isolasi bakteri endofit, karakterisasi morfologi, dan uji aktivitas antibakteri. Sebanyak 8 isolat bakteri endofit berhasil diisolasi dari daun ciplukan dan memiliki morfologi koloni yang bervariasi. Berdasarkan uji aktivitas antibakteri, semua isolat mampu menghambat pertumbuhan S.aureus dengan diameter zona hambat berkisar antara 0,5-2mm dan 2 isolat terhadap pertumbuhan bakteri E.coli dengan diameter sebesar 2mm.
At the beginning of 2020, the world was shocked by the emergence of a new disease outbreak, Covid-19, which was caused by the SARS COV-2 virus. The school environment is one of the places that is vulnerable to the spread of this virus. An effective preventive measure against the spread of Covid-19 is the application of clean and healthy living habits (PHBS) in the school environment, one of which is the habit of washing hands. However, not all residents in the school understand how to wash their hands properly. The purpose of this service activity is to provide education about proper hands washing according to WHO for MI Muhammadiyah Need in Magelang Regency. The material needed for the implementation of the activity is a hand sanitizer liquid made from 70% active alcohol as a hand disinfectant. Implementation of community service activities using demonstration methods and hands-on practice of hands washing according to WHO. Based on this service activity, not all members of the MI Muhammadiyah Need was known the steps for hands washing properly according to WHO. Through this service, school residents are given knowledge and practice the 8 steps of hand washing according to WHO using a handsanitizer.
