These results demonstrate that nitric oxide is an intermediate in angiotensin II stimulated TGF-beta1 expression but not in stretch induced TGF-beta expression, and that TGF-beta1 is a negative regulator of nitric oxide in rat renal epithelial cells. The complex interaction of these cytokines may be a target for intervention in the fibrotic and apoptotic processes in the obstructed kidney.
A high-efficiency fire retardant composition was prepared with dicyandiamide, phosphoric acid, boric acid, borax, urea and magnesium sulfate and it was used to process veneers which were then to prepare the plywood. Meanwhile, heat release and smoke release from combustion of plywood were tested by a cone calorimeter, including heat release rate, mass loss rate, CO yield, CO2 yield and oxygen consumption. Results showed that the plywood with this fire retardant treatment had the better flame-retardant performance and smoke suppression effect as well as the stronger char-forming capability compared to plywood without fire retardant treatment. The average heat release rate, total heat release, average effective heat of combustion, total smoke release, CO yield and oxygen consumption of the plywood with fire retardant treatment were decreased by 63.72%, 91.94%, 53.70%, 76.81%, 84.99% and 91.86%, respectively. Moreover, the fire growth index of plywood treated by fire retardant was relatively low (3.454 kW·m-2·s-1) and it took longer time to reach the peak heat release rate, accompanied with slow fire spreading. The fire performance index was relatively high (0.136 s·m2·kW-1) and it took longer time to be ignited, thus leaving a long time for escaping at fire accidents. The fire hazard of plywood with fire retardant treatment was low, and its safety level was high.
Spinal cord injury (SCI) remains an intractable clinical challenge of neurosurgery, it can be divided into two stages: uncontrollable primary injury induced by mechanical damage and controllable secondary injury regulated by continuous cell death. The apoptosis was the one of most important events in secondary injury, previous studies revealed that excessive endoplasmic reticulum (ER) stress breaks down the homeostasis and triggers apoptosis in the spinal cord. To deter or alleviate the secondary jury, we screen one of fat-soluble compounds, salubrinal, which was an inhibitor of eIF2α dephosphorylation can repair SCI by inhibiting ER stress in mice after SCI. Administration of salubrinal effectively represses apoptosis, protects neuronal cell, and promotes the restoration of locomotor function in mice SCI models. Furthermore, the level of phosphorylated eIF2α was raised in the presence of salubrinal, but the protein expression of ATF4 and CHOP was downregulated. Unexpectedly, transcriptional expression of CHOPregulated pro-apoptotic genes was decreased. These data suggest salubrinal suppress ER stress by targeting eIF2α/ ATF4 pathways and reduces cell death after SCI. It is suggested that the mitigation of secondary lesion by inhibiting ER stress induced apoptosis in the early phase of SCI is promising treatment strategy.
Objective: Short-term intensive insulin therapy (SIIT) can induce remission of type 2 diabetes (T2D) , and the improvement of insulin sensitivity is considered essential. This study aimed to evaluate the role of adiponectin change during SIIT in patients with newly diagnosed T2D. Methods: SIIT was given to 42 patients with newly diagnosed T2D to normalize blood glucose (4.4-6.0 mmol/L for fasting blood glucose [FBG], 4.4-8.0 mmol/L for 2h-postprandial blood glucose [2hBG]) for two weeks. Serum lipids, adiponectin, and high-sensitive C reactive protein levels, insulin resistance and β-cell function were evaluated before and after SIIT. Results:After the therepy, FBG (11.8±2.9 mmol/L vs. 6.8±1.3mmol/L,P<0.001) and 2hPG (18.4mmol/L± 6.4mmol/L vs. 9.7±2.4mmol/L, P<0.001) were both significantly decreased, with β-cell function (measured by acute insulin response [AIR], HOM-B) and insulin sensitivity (measured by HOMA-IR) improved. Serum adiponectin raised from 2.9±1.2mg/L to 3.4±1.1mg/L (P<0.001) ,while serum triglyceride and LDL-C decreased significantly. Serum adiponectin after treatment was correlated with baseline triglyceride (r = -0.33, P = 0.04) , HDL-C (r = 0.34, P = 0.03) , LDL-C (r = -0.31, P = 0.05) and Hs-CRP (r = -0.33, P = 0.05) ; the change of adiponectin (Δadiponectin) after treatment were significantly correlated with the level of Δtriglyceride (r = 0.38, P = 0.03) . There was no notable correlation between adiponectin and β-cell function index or HOMA-IR. Adiponectin levels were similar between patients who obtained diabetes remission (defined as FBG< 7.0mmol/L, 2hBG<10.0mmol/L without hypoglycemic agents) and those who did not (baseline:3.0±1.5mg/L vs. 2.9±1.0 mg/L; after SIIT:3.4±1.0mg/L vs. 3.3±0.9 mg/L,both P>0.05) . Conclusion: Adiponectin level was significantly increased after SIIT in patients with newly diagnosed T2D.
