BackgroundLignocellulose is the most abundant biomass on earth. However, biomass recalcitrance has become a major factor affecting biofuel production. Although cellulose crystallinity significantly influences biomass saccharification, little is known about the impact of three major wall polymers on cellulose crystallization. In this study, we selected six typical pairs of Miscanthus samples that presented different cell wall compositions, and then compared their cellulose crystallinity and biomass digestibility after various chemical pretreatments.ResultsA Miscanthus sample with a high hemicelluloses level was determined to have a relatively low cellulose crystallinity index (CrI) and enhanced biomass digestibility at similar rates after pretreatments of NaOH and H2SO4 with three concentrations. By contrast, a Miscanthus sample with a high cellulose or lignin level showed increased CrI and low biomass saccharification, particularly after H2SO4 pretreatment. Correlation analysis revealed that the cellulose CrI negatively affected biomass digestion. Increased hemicelluloses level by 25% or decreased cellulose and lignin contents by 31% and 37% were also found to result in increased hexose yields by 1.3-times to 2.2-times released from enzymatic hydrolysis after NaOH or H2SO4 pretreatments. The findings indicated that hemicelluloses were the dominant and positive factor, whereas cellulose and lignin had synergistic and negative effects on biomass digestibility.ConclusionsUsing six pairs of Miscanthus samples with different cell wall compositions, hemicelluloses were revealed to be the dominant factor that positively determined biomass digestibility after pretreatments with NaOH or H2SO4 by negatively affecting cellulose crystallinity. The results suggested potential approaches to the genetic modifications of bioenergy crops.
Two-year old seedlings of Silver buffaloberry (Shepherdia argentea (Pursh) Nutt.) were exposed to NaCl salinity (0, 200, 400 and 600 mmol/l) for 30 days. Leaf water potential (Ψ w ), chlorophyll contents (Chl a, b, and a + b) and K + content decreased with an increase in salinity. Relative water content (RWC) declined significantly with 400 and 600 mmol/l NaCl. However, salinity induced an excessive accumulation of Na + in the leaves of plants. Light responses of photosynthesis showed that net photosynthetic rate (P N ) values were continuously raised with the increase of photosynthetic photon flux density (PPFD) at all salinity levels and plants treated with 600 mmol/l salinity suffered from photoinhibition with the lowest P N values. The reduction of P N and stomatal conductance (g s ) associated with a sharp increase of intercellular CO 2 concentration (C i ) in the leaves at 600 mmol/l salt-treated plants showed that non-stomatal limitations might have prevailed over stomatal limitations under severe saline conditions, due to severe cellular dehydration, inhibited synthesis of chlorophyll and ionic imbalance and toxicity. It is concluded that S. argentea possesses high salt tolerance capacity and can be widely cultivated in salt-affected areas.
Th17 and regulatory lymphocyte subsets such as Tregs and Bregs have been reported to play important roles in autoimmune diseases. The aim of this work was to perform quantitative studies of circulating Th17, Tregs, and Bregs in patients with new-onset Graves' disease (GD). Twenty GD patients and 20 healthy controls were involved in this study. Blood samples were taken for flow cytometry detection of CD4+IL-17+ Th17, CD4+Foxp3+ Tregs, and CD19+CD1dhiCD5+ Bregs and meanwhile, for real-time PCR measurement of gene expressions of RORγt, IL-17 and IL-10. The proportions of Tregs and Bregs as well as the Foxp3 gene expression but not IL-10 were significantly decreased in GD group compared with the healthy controls. The frequency of Th17 together with the gene expressions of RORγt and IL-17 were significantly increased in the GD group. Furthermore, the Th17/Treg ratio was also significantly higher in GD group. A significant positive correlation between Th17 and TSAb (r = 0.656, p < 0.001) but significant negative correlations between Treg/Breg and TSAb (r = −0.339, p = 0.032; r = −0.759, p < 0.001) were identified among the participants. This study indicated that increased Th17 and impaired Treg responses, along with a decreased number of CD19+CD1dhiCD5+ Breg cells, were involved in GD pathogenesis.
