Air tebu dapat dimanfaatkan sebagai pengencer semen karena mengandung berbagai nutrien yang dapat menunjang kehidupan spermatozoa selama preservasi. Penelitian ini bertujuan untuk menguji kemampuan air tebu dalam mempertahankan daya hidup spermatozoa kambing PE selama preservasi pada suhu 3-5oC. Semen ditampung menggunakan vagina buatan dan segera dievaluasi secara makroskopis dan mikroskopis. Semen segar dibagi ke dalam lima buah tabung reaksi kemudain masing-masing diencerkan dengan 80% pengencer dasar laktosa + 20% kuning telur sebagai kontrol (P0), 20% air tebu + 60% akuabidetislata + 20% kuning telur (P1), 30% air tebu + 50% akuabidestilata + 20% kuning telur (P2), 40% air tebu + 40% akuabidestilata + 20% kuning telur (P3), dan 50% air tebu + 30% akuabidestilata + 20% kuning telur (P4). Semen yang telah diencerkan disimpan di dalam refrigerator lemari es pada suhu 3-5oC. Motilitas dan daya hidup spermatozoa masing-masing perlakuan dievaluasi setiap hari hingga motilitas mencapai 40%. Hasil penelitian diperoleh karakteristik semen segar kambing PE yaitu volume 1,15 ml, konsentrasi 3.870 juta/ml, motilitas 80%, spermatozoa hidup 87,75%, dan spermatozoa abnormal 7,5%. Persentase motilitas dan spermatozoa hidup pada hari keempat preservasi perlakuan P2 (41,25% dan 50%) nyata (P<0,05) lebih tinggi dibandingkan dengan P0 (37,5% dan 47%), P1 (32,5% dan 44%), P3 (30% dan 36,25%), dan P4 (18% dan 29,25%). Dapat disimpulkan bahwa Kombinasi pengencer terbaik untuk semen kambing PE adalah terdiri atas 30% air tebu + 50% akuabidestilata + 20% kuning telur. ABSTRACT Sugarcane juice can be used as an alternative semen extender because it contains various nutrients that can support the life of spermatozoa during preservation. This study aimed to examine the ability of sugarcane juice to maintain the ability of Etawah crossbreed goat spermatozoa during preservation at 3-5oC. Semen was collected using an artificial vagina and immediately evaluated macroscopically and microscopically. Fresh semen was divided into five tubes and diluted with 80% lactose-based extender + 20% egg yolk as a control (T0), 20% sugarcane juice + 60% aquabidestilata + 20% egg yolk (T1), 30% sugarcane juice + 50% aquabidestilata + 20% egg yolk (T2), 40% sugarcane juice + 40% aquabidestilata + 20% egg yolk (T3), and 50% sugarcane juice + 30% aquabidestilata + 20% egg yolk (T4), respectively. The diluted-semen was stored in the refrigerator at 3-5oC. The motility and viability of spermatozoa were evaluated every day until the motility reached 40%. The results showed that the characteristics of fresh semen of Etawah crossbreed goat were volume 1.15 ml volume, spermatozoa concentration 3870 million cells/ml, spermatozoa motility 80%, live spermatozoa 87.75%, and abnormal spermatozoa 7.5%. The percentage of motility and live spermatozoa on the fourth day of preservation for T2 (41.25% and 50%) was significantly (P<0.05) higher than T0 (37.5% and 47%), T1 (32.5% and 44%), T3 (30% and 36.25%), and T4 (18% and 29.25%). It can be concluded that the best extender combination for Etawah crossbreed goat semen is composed of 30% sugarcane juice + 50% aquabidestilata + 20% egg yolk.
