The progression of tumor depends on the level of Bcl-2 which protects the cells from undergoing apoptosis, and its level should be more than the level of the death inducer. So, our Aim: To investigate chemopreventive effect of olive oil in epithelial alteration which induced in hamster buccal pouch.Methods: Epithelial dysplasia will occur by the application of DMBA on alternative days for 8 weeks on hamster buccal pouch. Hematoxylin and Eosin (H&E) and Streptavidin-biotin immunoperoxidase staining techniques were used to detect the expression of Bcl-2 in three groups (G1 negative control or normal group, G2 DMBA group and G3 olive oil chemoprevention group). Evaluation of the immunostaining was done by using computer image analyzer system. Results:H & E stained histological section in G1 with normal mucosa of HBP intact, and continuous epithelium of thin keratinized stratified squamous epithelium while in G2 there were incidence of severe epithelial dysplasia with hyperkeratosis & G3 reduced dysplasia incidence and severity as compared to G2. Bcl-2 immunohistochemical expression in G1 in basal & suprabasal layers while in G2 & G3 located in all epithelial layers with statistically highly significant difference (P<0.01) in means values of Bcl-2 expression among (G2>G1, G3) and (G3>G1). Conclusion:Olive oil plays an important role in reduction and prevention of transformation of normal oral epithelium into epithelial dysplasia. The liquid form of olive oil may be considered as a new line of chemoprevention in oral dysplasia and the Bcl-2 expression evidenced this reduction of abnormality in epithelial dysplasia.
The aim of the present study was directed to investigate the effect of Glutathione (GSH) and Selenium (Se) as chemopreventive agents in experimentally induced hamster buccal pouch (HBP) epithelial dysplasia.Material and methods: Twenty five golden Syrian male hamsters were used as experimental animals and divided into five groups (G1, G2, G3, G4 and G5): G1: 5 animals were left untreated. G2: 5 animals, their right buccal pouches were painted with 0.5% 7, 12 dimethylbenz [a] anthracene (DMBA) 3 times a week for 8 weeks. G3: 5 animals were received GSH one week before, as well as during the application of DMPA for 8 weeks. G4: 5 animals were received Se one week before, as well as during the application of DMPA for 8 weeks. G5: 5 animals were received GSH and Se one week before, as well as during the application of DMPA for 8 weeks. After termination of the experiment, all animals were sacrificed, and the buccal mucosa was excised and preparation in order to be examined histologically and immunohistochemically, then statistical analysis based on these examinations was done Results: Regard to expression of Bcl-2, G2 had recorded the highest mean area percentage (44.25%), while G1 had recorded the lowest mean area percentage (6.73%) and there was high significant difference between G1,G2, G3, G4 and G5 where P value was <0.001, while in regard to expression of Bak, G1 had recorded the highest mean area percentage (41.02%), while G2 had recorded the lowest mean area percentage (19.79%) and there was significant difference between G1, G2, G3, G4 and G5 where P value was 0.019. Conclusion:GSH and Se are considered as promising chemotherapeutic agents in prevention of induced HBP epithelial dysplasia and proved important role in apoptosis and proliferation throughout the process of carcinogenesis.
Aim: To investigate the expression of p63 protein and ki67 antigen in a series of graded OSCC to evaluate the potential association between the expression of the two markers and the aggressiveness of histologic grades of OSCC. Methods: Hematoxylin and Eosin (H&E) and streptavidin-biotin immunoperoxidase staining techniques was used to detect the expression of p63 & ki67 in different grades of OSCC. Evaluation of immunostaining was done using computer image analyzer system & statistical analysis. Results: regarding p63 & ki67 expression were obtained by comparing the area % between different grading of OSSC, there was a statistically highly significant difference between different grades of OSCC (P<0.01) & there was a statistically significant direct correlation between p63 & ki67 stain. Conclusion: These results suggest that p63 & ki67 could reflect biological features, such as proliferation and differentiation for recognition of clinical behavior of OSCC also p63 & ki67 alteration may be associated with the progression of these neoplasms.
