SDS-agarose FN immunoblotting of 257 normal and pathological human plasma samples revealed the ladder pattern of multiple plasma FN bands which corresponded to FN monomer and dimer, and 5 FN-fibrin bands with increasing molecular masses. The FN-fibrin bands of about 750 kDa, 1000 kDa, 1300 kDa, 1600 kDa, and 1900 kDa appeared more frequently and in significantly higher relative amounts in the pathological samples (P < 0.000) than in relatively healthy individuals. The revealing of high-molecular FN-fibrin complexes by SDS-agarose FN immunobloting might have the potential to become a laboratory biomarker of some diseases in which the coagulation system is triggered.
The influence of the co-factor pyridoxine, vitamin B6, on the activity of aromatic amino acid decarboxylase enzyme was studied by positron emission tomography, PET in the brain of the Rhesus monkey using the precursor for serotonin synthesis 5-hydroxy-L-tryptophan (5-HTP) radiolabelled with 11C in the beta-position. The rate constant for the formation of serotonin in the corpus striatum was calculated using a two tissue compartment model with reference area in the brain. In baseline investigations, the mean rate constants (+/-S.D:) for selective utilization of [11C]5-HTP to form [11C]serotonin in the corpus striatum was 0.0080 +/- 0.0011 min(-1). Pretreatment with intravenous pyridoxine hydrochloride 10 mg/kg bodyweight before doing a second PET study resulted in an enhanced rate constant by a mean of 20%. The rate increase was statistically significant. The increase varied considerably in different monkeys from no effect to more than 60%. The effect of pyridoxine on aromatic amino acid decarboxylase activity supported a regulatory role of pyridoxine on the synthesis of neurotransmitter in vivo, and may be of importance in diseases with deficiencies in neurotransmitter function.
Two different aspects should be taken into consideration in HD patients treated with rHuEpo and concomitant i.v. iron therapy: (1) response of the erythropoietic system to rHuEpo, and (2) adequate delivery of the supplemented iron to the erythropoietic system. The patient's percentage of hypochromic RBC and increase in serum ferritin after i.v. iron supplementation should be used to decide whether or not i.v. iron should be given and to monitor this type of therapy in HD patients.
The aim of the study was to assess the acceptance level of COVID-19 vaccination among healthcare workers (HCW) and the general population in Poland at the start of the national COVID-19 vaccination program from 18–31 December 2020. A cross-sectional anonymous survey was conducted in a group of 1976 people: 1042 health professionals and 934 non-medical professionals using an on-line questionnaire. The most skeptical about the COVID-19 vaccine were students of non-medical faculties, non-medical professions, and administrative–technical health service staff (26.2%, 38.7% and 41.2%, respectively). The most positive attitude to vaccination was reported by doctors, medical students and pharmacists (80.6%, 76.9% and 65.7%, respectively). Doctors (64.7%) and medical students (63.7%) most often declared confidence in vaccines compared to nurses (34.5%). Distrust about vaccine safety was declared by nurses (46.6%) and pharmacists (40.0%). HCW encouraged others to vaccinate more eagerly (65.8%) than non-medical professions (28.3%). Thus, a considerable proportion of HCW in Poland expressed concern about vaccines just prior to the beginning of the COVID-19 immunization program. The significant decrease in the willingness to vaccinate observed in Poland towards the end of 2021 must be considered in the light of the serious COVID-19 vaccination hesitancy in the Polish population.
in the elderly, a division of patients into two groups shall be considered. The first group shall include patients with a longstanding asthma, whose symptoms occurred before the age of 65, and the second group shall include patients, in whom the symptoms occurred after the age of 65. It is worth mentioning a historical study by Lee and Stretton [8] on patients with late-onset (after the age of 65), severe bronchial asthma. Attention should, however, be paid to the fact that the study involved a small, 15-person group, mostly smokers complaining of a long-standing productive cough. In the study by Quadrelli et al. [9], the elderly with diagnosed late-onset asthma had similar intensity of disease symptoms to the young patients. Patients with a long-standing disease, contrary to the late-onset asthma patients, had a shorter symptom-free period, increased number of hospitalizations and emergency medical interventions during a year, as well as lower values of ventilation parameters. Braman et al. [10] observed that among elderly asthma patients, subjects with a long-standing disease have considerably impaired lung function, which results in symptoms similar to the ones observed in a chronic obstructive pulmonary disease (COPD). Contrary to the aforementioned researchers, Burrows et al. [11] did not found relations between the disease duration and its severity. Those authors showed that the course of bronchial asthma in the elderly patients, despite considerable ventilation impairment, in long-term follow-up is not characterized by a rapid deterioration. The observed discrepancies show that elderly patients diagnosed with bronchial asthma are a composite group. Additional difficulties occurring during attempts to characterize bronchial asthma in elderly patients result from the fact that the changes due to advanced age of studied persons overlap with the disease manifestations.
