Background: Specificity of G-protein function may be determined by a specific ␣␥ composition. Results: Combinatorial disruption of ␥ 3 and ␥ 7 produces a severe seizure phenotype not observed with either gene alone. Conclusion: This reflects distinct roles for ␥ 3 and ␥ 7 in G i/o -and G olf -signaling pathways that modulate seizure susceptibility. Significance: The ␥ subunits direct the assembly of distinct G-protein ␣␥ heterotrimers that specify diverse receptor actions.
