A micropropagation protocol was developed for multiplication of seedless lemon (Citrus limon L. cv. Kaghzi Kalan) using nodal explants. The maximum shoot regeneration was observed on low level of BAP (0.1 mg l(-1)) or kinetin (0.5 mg l(-1)). BA was recorded to be better than kinetin in terms of number of days taken to bud break. The maximum number of shoots per explant was observed on 0.1 mg l(-1) BA and 0.5 mg l(-1) kinetin. Shoot proliferation decreased with increasing concentration of BA alone, but in case of a combination of BA and NAA (0.1 mg l(-1) each), it increased with increasing concentration of BA up to 10.0 mg l(-1). None of the treatments including BA or kinetin alone or BA in combination with NAA produced significantly more shoots for commercial exploitation. In the case of a combination of BA + kinetin + IBA, the maximum (5.5 shoots per explants) proliferation was observed on MS medium containing 1.0 mg l(-1) BA + 0.5 mg l(-1) kinetin + 0.5 mg l(-1) IBA or 0.25 mg l(-1) BA + 1.0 mg l(-1) kinetin + 1.0 mg l(-1) IBA. Regenerated shoots showed root induction on MS basal medium or on MS medium containing 1.0 mg l(-1) IBA. It is concluded that a five-fold increase (1.0 mg l(-1) BA + 0.5 mg l(-1) kinetin + 0.5 mg l(-1) IBA) in axillary shoot proliferation, while seven-fold increase (0.25 mg/l mg l(-1) BA + 1.0 mg l(-1) kinetin + 1.0 mg l(-1) IBA) during the second cycle of multiplication could be obtained using the two plant growth regulator combinations. PCR amplification with 14 different random primers confirmed no somaclonal variant up to two cycles of shoot multiplication.
Five rice varieties viz. Binadhan-4, Binadhan-5, Binadhan-6, Binadhan-10 and Iratom-24 were evaluated in vitro under different water stress conditions. Several parameters such as germination percentage, shoot length, root length, shoot-root ratio, fresh weight, dry weight, turgid weight, relative water content and proline accumulation were studied. Drought condition was created by MS medium supplemented with five treatments of PEG, with a control such as 0%, 1%, 2%, 3% and 4% of PEG. The highest germination (100%) was found in the variety Binadhan-10 under low water stress conditions induced by 1% PEG. Similarly, the highest percentage of germination was found in all varieties under control condition (0% PEG). The lowest percentage of germination was obtained in the variety Iratom-24. But under severe stress (4% PEG), the highest percentage of germination was found only in the variety Binadhan-10. Moreover, the variety Binadhan-10 was found to be the best at 4% PEG for shoot length, root length, shoot-root ratio, relative water content and also the best at 1% PEG for fresh weight, dry weight, turgid weight. Water stress decreased relative water content and increased proline accumulation in rice. The highest relative water content was recorded in the variety Binadhan-10 and the lowest value recorded in the variety Binadhan-5. The highest proline content was obtained from the binadhan-6 at the highest treatment (4% PEG). Binadhan-10 showed the best performance almost in all the parameters under drought stress because of its own nature of tolerancy.Progressive Agriculture 27 (2): 128-135, 2016
Plant growth regulators (PGRs) namely, 2,4-D, NAA and BAP were added into Murashige and Skoog (MS) medium to observe the effect of PGRs on the growth and development of Dendrobium sp. orchid. Leaf tips of Dendrobium sp. were used as explants and inoculated on MS medium supplemented with 2, 4 D (0, 0.5, 2.5, 5, 10 mgL −1 ) for development of PLBs. The maximum PLBs formation (90%) and the maximum number of PLBs (16.00) were observed in 10 mgL −1 2, 4-D into MS medium after 60 days of culture. Subcultured PLBs were inoculated on MS medium supplemented with different combinations of NAA (0, 0.5, 2.5, 5 mgL −1 ) and BAP (0, 0.5, 2.5, 5 mgL −1 ) for shoot regeneration. The maximum number of shoot (11.00), the highest fresh weight (0.6233g) and the highest shoot length (3.613 cm) were observed in 0.5 mgL −1 NAA + 0.5 mgL −1 BAP after 60 days of culture. Even, the maximum number of root (4.00), the maximum root length (1.627cm) and the maximum plantlet regeneration percentage (93.33%) were observed with the combined effect of 0.5 mg NAA and 0.5 mg BAP after 60 days of culture. Finally, regenerated plantlets were transferred into half strength MS medium to obtain plants.
