Liver damage is a common problem all over the world. To optimize the drug provision in the limited financing of the Health Care System, it is necessary to develop new drugs with hepatoprotective properties that are highly effective and of low cost. That is why we developed the original drug Lavaflam. Lavaflam is a combination of the following substances: dry concentrate of Helichrysum arenarium and Lavender oil. The purpose of the research was to assess the biochemical evidence of the hepatoprotective properties of the drug Lavaflam on the experimental model of subchronic hepatitis in rats. Construction of an experimental model of subchronic hepatitis in rats, comparison of hepatoprotective properties of Lavaflam and Carsil preparations on the basis of biochemical research. The subject of the study was to determine the hepatoprotective properties of Lavaflam tablets. Experimental subchronic hepatitis induced in rats by way of intragastrical introduction of tetrachlormethane. The study drug Lavaflam in this experimental model of subchronic tetrachlormethane hepatitis in rats showed a positive effect on oxidant processes by increasing the compensatory mechanisms of antioxidant systems; inhibiting free radical pathology; having positive effect on the processes of biliary excretion, cholestasis, reduction of infiltrative, destructive and inflammatory process in the liver; decreasing the cytolytic process; restoring the structure of the membrane components of hepatocytes; stabilizing and enhancing functional activity of the liver; restoring its protein-synthetic function; and increasing the ability of the drug Lavaflam to restore metabolic and liver damage.As a result of the performed biochemical study of the hepatoprotective action of Lavaflam, it has been found that in the developed drug Lavaflam, the level of hepatoprotective action corresponds with the reference drug Carsil.
Biliyer sistem hastalıklarının tedavisi, modern tıbbın acil problemlerinden biridir. Gereç ve Yöntemler: Biliyer sistem hastalıklarının karmaşık tedavisi için, bitkisel bileşenler flamin (0.05 g) ve lavanta yağından (0.02 g) oluşan bir bileşimi içeren kombine tablet formundaki formdaki orijinal farmasötik ilaç "Lavaflam" önerilmiştir. Flamin, flavonoidler grubundan (salipurposid, isosalipurposide, kaempferol, luteolin) aktif maddelerden oluşan bir kompleks içeren, ölmez çiçeklerden [Helichrysum arenarium (L.) Moench, Asteraceae] elde edilen bitkisel kaynaklı ilaçtır. Kolesistit, kolanjit ve biliyer diskinezide koloretik ve anti-inflamatuvar ajan olarak uygulanır. Tabletler, ayrı granülasyon teknolojisi için preslenerek hazırlandı. Bulgular: Lavanta yağı granülesi katı faz metodu kullanılarak hazırlandı ve β-siklodekstrin yardımcı madde olarak kullanıldı. Flamin granülesi küresel şekilli dolgu maddesi mannitol PARTECK M 200 ile karıştırılarak hazırlandı. Önceki çalışmalara dayanarak, tasarlanan "Lavaflam" tablet bileşiminin eksipiyanları, β-siklodekstrin (0.27 g), mannitol PARTECK M 200 (0.20 g), kroskarmeloz sodyum (0.03 g), patates nişastası (0.022 g), PEG 6000 (0.002 g) ve magnezyum stearat (0.006 g) idi. Lavanta yağının ana bileşenlerinin analizi linalol ve linalil asetat referansları ile, gaz kromatografisi kullanılarak gerçekleştirildi. Flaminin toplam flavonoidlerinin analizi, referans olarak isosalipurposid ile spektrofotometri kullanılarak gerçekleştirildi. Sonuç: Yeni bitkisel kaynaklı tabletler "Lavaflam", Avrupa Farmakopesi gereksinimlerini aşağıdaki parametrelerle karşılamaktadır: görünüm, geometrik boyut, ortalama ağırlık, dağılma, ufalanabilirlik, tabletlerin kırılmaya karşı direnci ve miktar tayini. Materials and Methods: An original pharmaceutical drug "Lavaflam" in the form of combined tablets, which includes a composition of herbal components flamin (0.05 g) and lavender oil (0.02 g), was proposed for the complex treatment of diseases of the biliary system. Flamin is phytomedicine from the immortelle flowers [Helichrysum arenarium (L.) Moench, Asteraceae], which contains a complex of active substances from the flavonoids group (salipurposide, isosalipurposide, kaempferol, luteolin). It is applied as a choleretic and an anti-inflammatory agent in cholecystitis, cholangitis, and biliary dyskinesia. Tablets were prepared by pressing for separate granulation technology. Results: The lavender oil granulate was prepared using the solid phase method, and β-cyclodextrin was used as an excipient substance. The flamin granulate was prepared by mixing with the spherical-shaped filler mannitol PARTECK M 200. Hepatobiliary system diseases are medical and social problem at the present time. Important issue is the development of a new complex of herbal medicine and its pharmacoeconomic study.The purpose of the work was an experimental substantiation of the hepatoprotective properties of the drug Lavaflam.Methods of research: pharmacological, morphological, histological, methods of mathematical statistics. Materials of research.Experimental pharmacological studies were performed on the white rats weighing 180-220 g. The study was carried out in the Central Research Laboratory (CRL) of the National University of Pharmacy, Kharkіv, Ukraine (Certificate № 023/13 of 05.03.2013) according to the methodological recommendations of the State Expert Center of the Ministry of Health of Ukraine. The rats were divided into four experimental groups: the first group -intact control; the second grouppositive pathology -the animals were injected TChM; the third group -animals, received TChM and Lavaflam; the fourth group -animals, received TChM and reference drug Сarsil.Results. It has been found that the test drug Lavaflam showed hepatoprotective properties in experimental model of the subchronic hepatitis (diffuse inflammatory liver disease) in rats. Conclusions.Lavaflam, in comparison with Carsil contributed to the elimination and reduction in a number of pathological changes, increased adaptive capacity of hepatocytes, and Lavaflam promoted the physiological regeneration of cells. Relating to the effect of a positive influence on the hepatic parenchyma condition with induced subchronic hepatitis by TChM, the drug Lavaflam was not inferior to the comparison drug Carsil (by the effect on necrotic manifestations), or was superior to the comparison drug Carsil (by the zones of destruction spread, dystrophic changes in hepatocytes).
