Alternative medicine is used widely because there are a large health care alternatives to be more congruent within the own values and beliefs towards health. Alternative medicine is more compatible with patients, offers more personal autonomy and control over the health care decisions. It is accepted worldwide because of its compatibility and acceptability in increasing the beliefs regarding the nature and meaning of health and illness. This review deals with various alternative therapies used such as Ayurveda, homeopathy, acupuncture, naturopathy, yoga, herbal medicine, massage therapy; effectiveness of the alternative medicine; assessment of the effectiveness of alternative medicine; sources of information about the alternative medicine; alternative medicine therapies in treatment of various diseases; perceived benefits of alternative medicine and thereby concluding with the increased level of acceptance of alternative medicine, its widespread use in various diseases and their treatment with various alternative therapies.
Objective: Ramelteon, is a sleep agent that selectively binds to the MT1 and MT2 receptors in the suprachiasmatic nucleus (SCN), instead of binding to GABAA receptors. In the present research work, the formulation of ramelteon targeted to colon by using various polymers developed. Methods: Colon-targeted tablets were prepared in two steps. Initially, core tablets were prepared and then the tablets were coated by using different pH dependent polymers. Ethylcellulose, Eudragit RLPO and L100 were used as enteric coating polymers. The precompression blend of all formulations was subjected to various flow property tests and all the formulations were passed the tests. The tablets were coated by using polymers and the coated tablets were subjected to physical characterization, drug content, in vitro drug release and kinetics of drug release. Results: Among all the formulations, F4 formulation was found to be optimized as it was retarded the drug release up to 18 h and showed maximum of 99.25% drug release. It followed the first-order kinetics mechanism. All the formulations having Korsmeyer-Peppas ‘n’ values are in the range of 0.540 to 0.818. Hence, it was concluded that the prepared formulations followed non-Fickian diffusion. Conclusion: An effective and stable remelteon colon targeted formulation developed for treating insomnia.
Biologics include vaccines, recombinant proteins, genes, synthetic tissues and viruses, which have emerged from molecular and cellular biology, with respect to unmet clinical needs and expanding indications. Hence it is important at this stage to outline the essential events that have led to the current level of interest in protein and peptide drug delivery systems. Polymers consisting of amino acids that are covalently linked by peptide bonds are known as Proteins and inturn peptides are small proteins composed of few dozen amino acids. Due to large molecular sizes of the proteins ranging from thousands to several millions atomic masses sizes; absorption through the epithelial barriers in the gastrointestinal tract is low. Moreover, proteins are rapidly degraded by digestive enzymes. As the bioavailability by oral route is poor they can be administered by other routes mainly by parenteral (IV, SC, IM) injections. The biological activity of proteins is strongly dependent on their molecular structure i.e 10 structure; the amino acid sequence, which ultimately dictates the non-covalent interactions and the 20i.e alpha helices and beta helices; the periodic spatial arrangement of the polypeptide chain backbone and 30; the 3-dimensional conformation of the whole molecule, including the positions of all amino acid side chains. Some proteins may consist of multiple peptide chains grouped together by non-covalent intermolecular interactions. The arrangement of these subunits relative to each other constitutes the 40structure. An alteration at any level of molecular structure leads to a change or loss of biological activity. The present review focuses on the development of various routes of drug administration, formulation as well as stability aspects of protein and peptide drug delivery system.
