A new set of pyrazolo[3,4-d]pyrimidine and pyrazolo[4,3-e][1,2,4]triazolo[1,5-c]pyrimidine scaffolds (4–13) as well as the thioglycoside derivatives (14, 15) were designed, and synthesized as novel CDK2 targeting compounds.
Background: Para-umbilical hernioplasty is mostly safe but there are possible complications like seroma, wound infection and recurrence. Tranexamic acid is a synthetic derivative of the amino acid lysine that exerts an anti-fibrinolytic action through the reversible blockade of lysine binding sites on plasminogen molecules, used in the prevention and treatment of excessive bleeding both in primary and secondary care. Our study aimed to evaluate the effect of intravenous tranexamic acid injection on reduction of seroma after para-umbilical hernioplasty.Methods: We conduct a prospective controlled trial to compare the effect of intra-operative, intravenous single dose of tranexamic acid by a dose of 10 mg/kg body weight with induction of anaesthesia on the occurrence of local wound complications as seroma, wound infection in addition to time of drain removal and the amounts of drain output, by comparison between two groups.Results: Single intravenous dose of tranexamic acid with induction of anathesia reduced drain duration (P-value was 0.001), drain output in 1st day (P-value was 0.008), and drain output in 5th day (P-value was 0.036). Adverse effects were not observed.There were no significant difference in occurence of seroma (P-value was 0.429), and wound infection (P-value was 1.00).Conclusions: Our study revealed that tranexamic acid showed significant correlation with decrease duration of drains and fewer amounts of drain volumes in first day post-operative.
Cyclin-dependent kinase inhibition is considered a promising target for cancer treatment for its crucial role in cell cycle regulation. Pyrazolo pyrimidine derivatives were well established for their antitumor activity via CDK2 inhibition. In this research, new series of pyrazolopyrimidine derivatives ( 4–15 ) was designed and synthesised as novel CDK2 inhibitors. The anti-proliferative activities against MCF-7, HCT-116, and HepG-2 were used to evaluate their anticancer activity as novel CDK2 inhibitors. Most of the compounds showed superior cytotoxic activity against MCF-7 and HCT-116 compared to Sorafenib. Only compounds 8 , 14 , and 15 showed potent activity against HepG-2. The CDK2/cyclin A2 enzyme inhibitory activity was tested for all synthesised compounds. Compound 15 showed the most significant inhibitory activity with IC 50 0.061 ± 0.003 µM. It exerted remarkable alteration in Pre G1 and S phase cell cycle progression and caused apoptosis in HCT cells. In addition, the normal cell line cytotoxicity for compound 15 was assigned revealing low cytotoxic results in normal cells rather than cancer cells. Molecular docking was achieved on the designed compounds and confirmed the two essential hydrogen binding with Leu83 in CDK2 active site. In silico ADMET studies and drug-likeness showed proper pharmacokinetic properties which helped in structure requirements prediction for the observed antitumor activity.
Background: Breast cancer is one of the main problems related to women's health, and the various forms of treatment can lead to bodily changes and significantly alter women's sexuality and psychological status. Aim: The aim of this study was to evaluate the effect of nursing counseling guided by BETTER model on sexuality, marital Satisfaction and psychological status among breast cancer women. Design: A quasi-experimental research design. Setting: The study was conducted at an Outpatient clinic, Oncology Institute, Menoufia University-Egypt. Sampling: A purposive sample, composed of 87 women was recruited in the study. Tools: Six tools were used for data collection a) A Structured interviewing questionnaire b) Female sexual function index (FSFI) c) Body image scale d) ENRICH marital satisfaction scale E) Tylor anxiety scale (Arabic version) f) Perceived stress scale (PSS). The results: This study showed that there was a highly significant difference at study participants sexuality, marital satisfaction and psychological status pre and post application of nursing counseling guided by BETTER model. Conclusion: Application of nursing counseling guided by BETTER model had high significant impact on improving sexuality, marital satisfaction and psychological status of breast cancer women undergoing treatment. Recommendation: Adopt a BETTER sexual model of counseling for management of sexual dysfunction and psychological problems in breast cancer institutions. Ongoing education for maternity nurses on the impact of breast cancer treatment on women's sexuality and psychological status, as well as strategies to address women's sexual and psychological problems.
One of the most common procedures among pregnant women is a cesarean section. The worry that pregnant women experience throughout the preparatory stages of operation day, as well as their lack of control over being in a new setting and feeling in danger, causes anxiety and, as a result, instability. The aim was to determine the effect of video-assisted teaching guidelines on knowledge regarding anxiety levels among primigravida mothers undergoing caesarian sections. Subjects and method: Design: A quasi-experimental research design was utilized to achieve the aim of this study. Setting: the research was conducted at the Antenatal Outpatient Clinic at South Valley University Hospital. Subjects: A purposive included 139 primigravida mothers who were included in the study within six months. Three tools were used: Tool (I) A structured interview questionnaire, Tool (II) Primigravida mothers' knowledge regarding caesarian section (pre/post), and Tool (III) Anxiety Rating Scale (pre/post). Results: The majority of primigravida mothers reported that the main source of information regarding their knowledge was nurses. There was a positive significant correlation (P=0.005) between primigravida mothers ' knowledge scores pre and post-video-assisted teaching guidelines. There were highly significant improvements in primigravida mothers' knowledge regarding the caesarian section post-video-assisted teaching guidelines (P=0.005). Statistical highly significant differences and reductions were detected between anxiety levels of primigravida mothers pre and post-video-assisted teaching guidelines. Conclusion: The present study concluded that video-assisted structured teaching guidelines had a highly significant positive effect on improving knowledge and reducing anxiety levels among primigravida mothers undergoing caesarian section. Recommendations: Educational guidelines about the caesarian section should be taught and discussed in the antenatal care follow-up visits for primigravida mothers.
