Being aware that some working cancer survivors may have negative work-related experiences and/or may not maintain full employment in later survivorship years may enable cancer survivors and employers to improve survivors' experiences at work.

Nucleophosmin(NPM1orB23.1)isaubiquitouslyexpressednuclearphosphoproteinthatplayskeyroleinseveralcellular processes,includingribosomebiogenesis,centrosomeduplication,cellcycleprogression,cellgrowth,andtransformation.NPM1is oneofthemostfrequentlymutatedgenesinAML.EDAGisahematopoietictissue-specifictranscriptionregulatorthatplaysakeyrole inmaintainingthehomeostasisofhematopoieticlineagecommitment.InAMLpatients,thehighexpressionlevelofEDAGis associatedwithpoorprognosis.OurpreviousstudysuggestthatEDAGisaphysiologicalbindingpartnerofNPM1andregulates NPM1proteinstability,however,whetherEDAGregulatesNPM1inAMLpatientsandwhetherEDAGregulatesNPM1mutations remainunknown.Inthepresentstudy,wefoundthatinbonemarrowCD34 + cellsfromAMLpatients,silencingofEDAGledto decreasedproteinstabilityofNPM1proteinandincreasedcellsensitivitytodaunorubicin.AlthoughEDAGfailedtointeractwith NPMc + andregulateitsproteinstabilityinnormalculturecondition,withleptomycinBtreatment,EDAGoverexpressionenhancedthe proteinstabilityofNPMc + .InAMLpatientswithNPMc + ,theCD34 + cellsweremoreresponsivetodaunorubicintreatmentthanthe cellsfromAMLwithwildtypeNPM1,andsilencingofEDAGweaklyincreasedthesensitivitytodaunorubicin.Theseresults suggestedapotentialroleofEDAGinchemotherapyofAMLandthe"escape"ofNPMc + proteinfromEDAGstabilizationmight contributetothefavorableprognosisofAMLwithNPMc + .
