In order to analyze the genetic diversity and phylogenetic status of the Korean Chikso breed, we determined sequences of mtDNA cytochrome b (cyt b) gene and performed phylogenetic analysis using 239 individuals from 5 Chikso populations. Five non-synonymous mutations of a total of 15 polymorphic sites were identified among 239 cyt b coding sequences. Thirteen haplotypes were defined, and haplotype diversity was 0.4709 ranging from 0.2577 to 0.6114. Thirty-five haplotypes (C1–C35) were classified among 9 Asia and 3 European breeds. C2 was a major haplotype that contained 206 sequences (64.6%) from all breeds used. C3–C13 haplotypes were Chikso-specific haplotypes. C1 and C2 haplotypes contained 80.5% of cyt b sequences of Hanwoo, Yanbian, Zaosheng and JB breeds. In phylogenetic analyses, the Chikso breed was contained into B. taurus lineage and was genetically more closely related to two Chinese breeds than to Korean brown cattle, Hanwoo. These results suggest that Chikso and Hanwoo have a genetic difference based on the mtDNA cyt b gene as well as their coat color, sufficient for classification as a separate breed.
In this study, we have compared the skeletal muscle proteome at various stages of porcine postnatal development.Korean native pigs were divided into five postnatal stages of 30, 70, 130, 170 and 300 d and their loin muscles were analyzed for muscle proteome by using two-dimensional electrophoresis and mass spectrometry. We found 5 proteins showing a consistent pattern during skeletal muscle growth. Four proteins were identified as myosin light chain 1 slow-twitch (MLC1sa) isoform, troponin T, triosephosphate isomerase (TIP) and DJ-1 protein. The remaining protein was not identified. Two muscle fiber proteins of MLC1sa isoform and troponin T showed a high expression level at an early postnatal stage and then their levels were decreased markedly during growth stages. On the other hand, the expression of TIP and DJ-1 protein, which are well known as catalysis enzyme and antioxidantrelated protein, respectively, were linearly increased during growth stages. Thus, the stage-related muscle proteins may be useful as parameters for understanding the developmental characteristics of biochemical and physiological properties in Korean native pig skeletal muscle.
The level of genetic variation and relationships in three native Korean goat populations (Dangjin, Jangsu, and Tongyeong) as well as the populations of a farm were analyzed, based on 30 microsatellite markers. A total of 277 distinct alleles were observed across the four goat populations, and 102 (36.8%) of these alleles were unique to only one population. The mean observed heterozygosity and polymorphism information content were calculated as 0.461~0.651 and 0.462~0.679, respectively. In the NJ tree constructed based on Nei's DA genetic distance, the four populations represented four distinct groups. However, the genetic distances between each Korean native goat population and the farm population were two times those among the three native Korean breeds. The genetic structure within the three Korean native goat populations was also investigated. Cluster analysis, using the STRUCTURE software, suggested three clusters. The molecular information of genetic diversity and relationships in this study will be useful for the evaluation, conservation, and utilization of native Korean goat breeds as genetic resources.
