Background: Small molecule MEK1/2 inhibitors are designed to block ERK1/2-mediated signaling and inhibit proliferation. Results: Novel instrumentation which measures cellular respiratory chain function found that four structurally distinct MEK1/2 inhibitors acutely affected mitochondrial bioenergetics.
Conclusion:The anti-mitochondrial effects of MEK1/2 inhibitors determined proliferative potential. Significance: Impaired mitochondrial metabolism following MEK1/2 inhibition must be considered when using small molecule inhibitors to define this signaling pathway.
