Black orchid (Coelogyne pandurata Lindley) is one of the rare species of Coelogyne Lindley, protected by the government of Indonesia. The cytological character of orchid is very important to study to support the success of breeding. The study aimed to assess the chromosomal number, karyotype pattern and ploidy level the hybrid of Black Orchid (Coelogyne pandurata). This study shows that the chromosomal number of the hybrid of Coelogyne pandurata (2n = 36) >< Coelogyne rumphii (2n = 72) is 2n = 54. Ploidy analysis by flow cytometry shows that hybrid shows triploid (2n = 3x = 54) different from the parent Coelogyne pandurata which is diploid (2n = 2x = 36) and the parent Coelogyne rumphii which is tetraploid (2n = 4x = 72). However, both parents and their hybrid performed the same karyotype pattern, which is metacentric. The chromosome size showed a variation in length from the longest Coelogyne pandurata 2.98 ± 0.15 µm to the shortest Coelogyne rumphii 2.24 ± 0.15 µm. The hybrids have a range of 2.50 ± 0,10 µm to 2.85 ± 0.10 µm. Karyotype patterns of black orchid (C. pandurata), C. rumphii and the hybrid is metacentric.
Black Rice is rice producing anthocyanin in high intensity and popularly consumed as functional food. Long harvesting age and low productivity are the inhibiting factors in black rice breeding. High plant crown potentially results in plant fall-down. Mutation induction is expected to be able to improve the character of black rice. Therefore, the objectives of this research were to get more early-ripening black rice mutant, with shorter plant crown and increase productivity by inducing mutation in three varieties of local black rice using gamma-ray radiation. This experiment employed three local varieties as the first factor comprised Cempo Ireng, Cempo Melik and Melik. The second factor was the gamma-ray radiation dose, consisting of 4 levels: without radiation, radiations at 100 Gy, 200 Gy and 300 Gy doses. The results indicated that Melik variety is very potential. Melik variety has shorter plant crown with a responsive character in number of tiller for higher productivity. Moreover, radiation significantly reduces the duration of flowering and harvesting. However, to achieve a more stable character and lower flowering and harvesting period, the radiation needs to be continued in the next generation. The doses of radiation produces a response that varies in both morphological and biochemical properties.
<p>Durian is a tropical plant which can easily be found in South East Asia country such as Indonesian. Ngrambe village which it is located on Ngawi regency is one of central durian in East of Java. This research aimed to obtain data related to quantity and variety of superior local durian based on the morphological character. This study was conducted at Giriharjo, Ngrambe, Ngawi from October 2015 to January 2016. Based on the field survey, it was found that there are 60 varieties consist of 36 local durians, 9 superior released durians, and 15 introduction durians. By selection process among 60 varieties durian, there was found that 6 of local durians was potential as a superior character to be released as new superior variety. The varieties are local Tugini, local Kasmadi 1, local Kasmadi 2, local 13, local Wasis and local Miyem.</p>
A better understanding of variability in plant populations is crucial for crop improvement which plant breeders can use through selection. This study aimed to determine selection criteria for the sixth generation of black rice through genetic variability, heritability value and genetic advance. This research was conducted from March to July 2018 at Pakahan Village, Jogonalan Sub-district, Klaten Regency, Central Java, using a Randomized Complete Block Design (RCBD). The variables observed were plant height, total number of tillers and productive tillers, panicle length, number of seeds per panicle, panicle intensity index, weight per panicle, 1,000 seed weight, seed weight per plant, flowering age, harvest age, plot yield, yield per hectare, rice color and total anthocyanin content. Variance analysis showed that there was a very significant difference (p <0.01) among the genotypes tested for all observed characters indicating variability. In this study, the phenotypic coefficient of variance (PCV) values was relatively higher than the genotypic coefficient of variance (GCV) for all traits. However, GCV values were close to PCV values in some characters such as plant height, flowering age, harvest age, rice color and anthocyanin content which showed a high contribution of genotypic effects to the phenotypic expression of these characters. The high heritability and high average value of genetic advance were indicated in the parameters of plant height, flowering age, harvest age, rice color and anthocyanin content. The characters of plant height, flowering age, harvest age, rice color and anthocyanin content can be used as sixth-generation black rice selection criteria.
<p>Banana is one of the favorite tropical fruit. Banana business development begins with the provision of quality seeds. Tissue culture is a seed propagation technology that can produce large amounts of seed and a relatively short time by utilizing limited plant material. The planlets result of tissue culture needs to be acclimatization before planted in the field. Hydroponic substrate-based acclimatization is expected to produce seeds that are free from soil pathogens. Adjustment of the concentration of nutrients is important in order to produce a growth response that significantly affects and does not cause poisoning or plasmolysis. This study aims to determine whether hydroponic substrate types and nutrient concentrations affect the growth of Raja Bulu Kuning Banana plantlets. The experiment was carried out at net house, Faculty of Agriculture, Universitas Sebelas Maret Surakarta in October 2016 - January 2017. The treatment factors were substrate types (bagasse, arenga peat, steamed husk) and concentration of the nutrient solution (equivalent to EC 1.8, EC 2.0, and EC 2.5 mScm-1). The result showed that steamed husk is the best as the hydroponic substrate, while nutrient solution concentration equivalent to EC 1.8 mScm-1 is sufficient for acclimatization of Banana plantlets cv.Raja Bulu Kuning.</p>
