Platelets store and release CXCL12 (SDF-1), which governs differentiation of hematopoietic progenitors into either endothelial or macrophage-foam cells. CXCL12 ligates CXCR4 and CXCR7 and regulates monocyte/macrophage functions. This study deciphers the relative contribution of CXCR4–CXCR7 in mediating the effects of platelet-derived CXCL12 on monocyte function, survival, and differentiation. CXCL12 and macrophage migration inhibitory factor (MIF) that ligate CXCR4–CXCR7 induced a dynamic bidirectional trafficking of the receptors, causing CXCR4 internalization and CXCR7 externalization during chemotaxis, thereby influencing relative receptor availability, unlike MCP-1. In vivo we found enhanced accumulation of platelets and platelet-macrophage co-aggregates in peritoneal fluid following induction of peritonitis in mice. The relative surface expression of CXCL12, CXCR4, and CXCR7 among infiltrated monocytes was also enhanced as compared with peripheral blood. Platelet-derived CXCL12 from collagen-adherent platelets and recombinant CXCL12 induced monocyte chemotaxis specifically through CXCR4 engagement. Adhesion of monocytes to immobilized CXCL12 and CXCL12-enriched activated platelet surface under static and dynamic arterial flow conditions were mediated primarily through CXCR7 and were counter-regulated by neutralizing platelet-derived CXCL12. Monocytes and culture-derived-M1–M2 macrophages phagocytosed platelets, with the phagocytic potential of culture-derived-M1 macrophages higher than M2 involving CXCR4–CXCR7 participation. CXCR7 was the primary receptor in promoting monocyte survival as exerted by platelet-derived CXCL12 against BH3-mimetic induced apoptosis (phosphatidylserine exposure, caspase-3 activation, loss of mitochondrial transmembrane potential). In co-culture experiments with platelets, monocytes predominantly differentiated into CD163+ macrophages, which was attenuated upon CXCL12 neutralization and CXCR4/CXCR7 blocking antibodies. Moreover, OxLDL uptake by platelets induced platelet apoptosis, like other platelet agonists TRAP and collagen-related peptide (CRP). CXCL12 facilitated phagocytosis of apoptotic platelets by monocytes and M1–M2 macrophages, also promoted their differentiation into foam cells via CXCR4 and CXCR7. Thus, platelet-derived CXCL12 could regulate monocyte-macrophage functions through differential engagement of CXCR4 and CXCR7, indicating an important role in inflammation at site of platelet accumulation.
Purpose The purpose of this paper is to review the advanced technologies and approaches for utilization of waste generated in dairy industry. Whey is highly contaminated, with a high organic load around 100,000 mg O2/L COD (chemical oxygen demand), and is not used for further processing. The waste generated in different food industries can be utilized in different value addition product with the help of advanced technology. Design/methodology/approach Major well-known bibliometric information sources are the Web of Science, Scopus, Mendeley and Google Scholar. Several keywords like nutrition value of whey, whey utilization, whey valorization, whey technologies, whey beverages, fruit-based whey beverage, carbonated beverage, probiotic or alcoholic beverages, herbal beverage, fermented beverage and current scenarios were chosen to obtain a large range of papers to be analyzed. A final inventory of 126 scientific sources was made after sorting and classifying them according to different criteria based on topic, academic field country of origin and year of publication. Findings The comprehensive review of different literature, data sources and research papers seeks to find and discuss various sustainable solutions to this huge waste generated from milk industry. The sustainable use of whey for production and conversion in different types of products can uplift the bio-based economy of industries and thereof national/international economy. The recent upsurge in consumer interest for health-promoting products has opened up new vistas for whey beverages and other whey products research and development. Originality/value The paper draws out different sustainable characteristics and technology of whey products available in market, as well as potential products to be launched in the market. Interestingly, over the past few years, dairy industries have applied various technologies to process cheese whey and are in search of new products which can be prepared from the by-product. This review discusses on the recent research development of whey valorization with particular reference to technologies used in the addition to their commercial availability and a way forward.
Food safety and quality has received attention in the agri‐food sector and is basis of all initiatives taken on different activity levels starting from farm to enterprises as a whole on regional, national, and international levels. A hybrid of the ISO 9001, Quality Management System and Hazard Analysis and Critical Control Point (HACCP), ISO 22000 has been developed as an international solution for improving the food safety. Instead of applying good manufacturing practices, HACCP and ISO 9001:2000 separately, ISO 22000:2005 is implemented to observe the synergetic effect and to ensure food safety in food supply chain. ISO 22000 also known as Food Safety Management System (FSMS) is an international auditable standard. Standard ensures safe food supply throughout the chain and provide a framework of internationally harmonized system for the global approach. ISO 22000 incorporates critical control point and hazard analysis systems in more improved form to produce much effective auditable FSMS. This standard endorses conformity of services and products for international trading by assuring about reliability, food quality, and food safety.Practical applicationsThe role exploration of Food Safety Management System (FSMS) in implementing food safety throughout the food production and supply chain is reviewed in this paper. The goal “once certified, accepted worldwide” of Globe Food Safety Initiative theme discussed to help industries and researchers. ISO 22000 along with its sister standards are auditable, reliable, and reasonable to ensure safe production, distribution, and consumption of food. During review of literature it is revealed that all technical aspects and requirements to implement the FSMS are not discussed on one place and thus leading to confusion. The aim of this review is to discuss different requirements, documentations, and systems in place for the certification of standards and the article will be helpful for the industries, technocrats, academicians, researchers, and policy framers.
The purpose of present study was to determine the occurrence and etiology of mastitis in traditionally managed buffaloes. A total of 5707 quarter milk samples from 2057 buffaloes were examined. Of these, 2948 (51.65%) samples were found culturally positive. Among these, 1070 cases were from clinical mastitis and rest 1878 cases were positive for subclinical mastitis. As many as 3447 isolates were obtained from infected quarters. Out of these, 38.81% were Staphylococcus spp., 32.4% Streptococcus spp., 11.80% E. coli, 5.2% Corynebacterium spp., 1.36% Bacillus spp., 2.03% Klebsiella spp., 0.78% Pseudomonas aeruginosa, 0.14% Proteus , 0.14% yeast. Staphylococcus spp. was predominant mastitogenic organisms followed by Streptococcus spp. Of the staphylococcal organisms, Staphylococcus epidermidis was the most prevalent being present in the 63.15% of the isolates. Among streptococci, Streptococcus agalactiae were the predominant organisms followed by Streptococcus dysgalactiae and Streptococcus uberis. The mixed infections were detected in 7.33% quarters in different combinations. Most common combination was of Staphylococcus spp. and Streptococcus spp. followed by Streptococcus spp. and Corynebacterium spp. Results of Antibiotic sensitivity were variable
Aim:Anaplasma marginale is a rickettsial pathogen responsible for progressive anemia in ruminants leading to huge economic losses. The objectives of this cross-sectional study were to determine the prevalence of anaplasmosis and therapeutic evaluation of traditional line of treatment.Materials and Methods:A total of 168 cattle presented to Teaching Veterinary Clinical Complex, Lala Lajpat Rai University of Veterinary & Animal Sciences, Hisar during the period of 3 months (July-September, 2014) with history of fever, anorexia, reduced milk yield and tick infestation were analyzed for prevalence of hemoprotozoan diseases using classical giemsa stained thin blood smear parasitological method.Results:Out of these 168 animals, 7 (4.17%) were found to be suffering from anaplasmosis on the basis of presence of dense, rounded, intra-erythrocytic bodies situated on or near the margin of the erythrocytes. Overall prevalence of theileriosis and babesiosis were found to be 42.9% and 1.8%, respectively. Level of parasitemia was noticed to be 1.2%, 0.8% and 0.9% in babesiosis, theileriosis, and anaplasmosis, respectively. The most marked and common clinical signs reported in all the cases were severe anemia (hemoglobin=3-6 g/dl) and history of fever, followed by normal body temperature. Following treatment with oxytetracycline parenterally along with supportive therapy out of seven cases six got recovered without any side-effects.Conclusion:The current study indicates the emerging status of anaplasmosis in this part of the country as during the past few years there are very few reports showing the prevalence of clinical cases of anaplasmosis. Treatment with oxytetracycline yielded excellent result showing recovery in most of the clinical cases.
