Introduction: Wounds fatality is possible on instances where treatment is not conducted correctly. Moreover, Aloe Vera possesses anti-microbial activities, while ozone has bactericidal and antioxidant effects. This study is, therefore, aimed at assessing the number of neutrophils and the diameter of full-thickness wounds, following the multilevel dose administration of ozonated aloe vera oil.Methods: This is an experimental study, based on a post-test only control group design, with treatment and control groups. A total of 50 Sprague Dawley rats (250 ± 50 grams) were randomly divided into 10 units, the control was administered aloe vera oil, gentamicin ointment, and the treatment with ozonated aloe vera oil at varied dose of 600, 1200, and 1800 mg/ml. Subsequently, five groups were terminated on the 3rd day, while the remaining groups were euthanized on the 7th day. The neutrophil counts were obtained after histological examination with Hematoxylin Eosin (HE) stain, while the wound diameter was measured using calipers.Results: There were significant differences between the control and treatment groups. This was evidenced in the decreased neutrophil count after administering ozonated aloe vera oil at a dose of 1800 mg/ml, compared to 1200 and 600 mg/ml, and also aloe vera oil and gentamycin ointment. Furthermore, wound diameter was substantially reduced with all ozonated treatments, than the aloe vera oil and gentamycin treatments.Conclusion: Ozonated aloe vera oil is characterized by the capacity to improve wound healing
Background The 1st, 2nd and small 3rd degree hemorrhoids are non-operatively treated by using anti-inflammatory and plebotonic medicines, high fiber diet and changing lifestyle. Graptophyllum pictum (L.) Griff (GP) is potential to be a hemorrhoid medicine (anti-inflammatory and plebotonic medicine) since its anti-inflammatory effect may be related to antioxidant property. ObjectiveTo elaborate the anti-inflammatory and anti-oxidant effect of GP on experimental hemorrhoid in male Wistar rats. MethodsA Randomized Controlled Trial Post-test only design in hemorrhoid-like rats using croton oil induced into the anus for 30 seconds each day for 3 consecutive days. 14 Wistar rats were randomly allocated into 2 groups. Group 1 got normal saline solution, while group 2 treated with GPE 100mg/Kg bw for 5 consecutive days. On the 9th experimental day, the blood was drawn for Elisa examination of COX-2 and SOD levels. As COX-2 and SOD normally distributed, pool t-test and product moment correlation was used for statistical analysis. Results All of Wistar rats were still alive. COX-2 levels (ng/ml) in the GPE group was 266.28±158.47, was significantly lower compared to control (595.21±68.92) (p=0.000). Serum SOD levels (ng/ml) in the GPE was 77.00±11.0, and was significantly higher than the control 28.00±4.00 (p=0.02). Correlation between COX-2 and SOD serum level was significantly strong negative correlation (p=0,000 and r=-0.842) Conclusions GPE at a dose of 100 mg/kg bw has antioxidant and anti-inflammatory effects on artificial hemorrhoids through decreased COX-2 levels, and elevated SOD levels. Phycocyanin from Spirulina platensis extract has anticancer activity against various types of cancer cell cultures. However study about its effect on colon cancer cell lines, especially the WiDr, has not been reported before. This study aimed to reveal the anticancer activity of phycocyanin from Spirulina platensis extract on WiDr cells. The research was an in vitro experimental study, with the investigation on cytotoxicity also antiproliferation as the anticancer parameters. Both cytotoxicity and antiproliferation test was conducted through MTT assay to observe the visualization and inhibition of proliferation of different concentrations of phycocyanin in several incubation times on the WiDr colon cancer cell line. The obtained data were then processed statistically with the Two Way ANOVA test at a significance value of p <0.05 and followed with the Post Hoc test since there were significant differences. Based on the results, it could be postulated that phycocyanin extracted from freshwater Spirulina platensis was classified as non-toxic (IC50 of 855 µg/ml). Consequently, it is less potential to be used as the treatment for colon cancer. However, phycocyanin could inhibit the proliferation of the WiDr cell for approximately 47.4%, specifically at the concentration of 1710 µg/ml for 72 hours. It could be concluded that freshwater phycocyanin is less effective as an anticancer substance. The benefit of this study is to provide the new scientific evidence of the contrary results of freshwater phycocyanin activity from Spirulina platensis as an anticancer agent of colon cancer.
Latar belakang : Cisplatin dapat menyebabkan penurunan sistem hemopoetik akibat Radical oxygen spesies (ROS) pada penderita kanker kepala dan leher (KKL). Kombinasi vitamin C dan E dosis tinggi sebagai antioksidan dari luar diperlukan untuk menetralisir ROS. Tujuan penelitian ini adalah membuktikan bahwa kombinasi vitamin C dan E dosis tinggi dapat mengurangi penurunan sistem hemopoetik penderita KKL akibat cisplatin. Metode : Penelitian eksperimental pre-post test design. Subyek yang memenuhi kriteria inklusi dari klinik dan bangsal THT-KL RSUP Dr. Kariadi Semarang dilakukan randomisasi blok, kemudian dibagi menjadi dua kelompok; kelompok perlakuan diberikan kombinasi vitamin C 1000 mg dan E 400 mg dan kelompok kontrol diberikan vitamin C 2x50 mg selama 5 minggu. Analisis data dengan chi square, paired t-test dan independent t-test. Hasil : Empat puluh delapan subyek memenuhi kriteria inklusi, usia terbanyak 50–59 tahun yaitu 35,6%, laki-laki dan perempuan 3 : 1, KNF 32(71,1%), stadium IV 27(60%). Terdapat penurunan hemoglobin dan lekosit yang bermakna antara kedua kelompok (p<0,05), sedangkan penurunan eritrosit dan trombosit tidak berbeda bermakna (p>0,05) Simpulan : Kombinasi vitamin C dan E dosis tinggi dapat mengurangi penurunan kadar hemoglobin dan jumlah lekosit penderita KKL akibat cisplatin. Kata kunci: Kombinasi vitamin C dan E dosis tinggi, cisplatin, sistem hemopoetik.
