Objective: The objective of this study to investigate the effect of simvastatin on kidney fibrosis in mice with a 5/6 subtotal nephrectomy.Methods: Thirty adults (3 mo old) male Swiss mice were submitted to a 5/6 subtotal nephrectomy and studied after 14 d. Animals were divided into five groups: 5/6 subtotal nephrectomy (SN, n=6), sham operation (SH, n=6), simvastatin 5.2 mg/kg body weight (SIM-1, n=6), simvastatin 10.4 mg/kg body weight (SIM-2, n=6), and simvastatin 20.8 mg/kg body weight (SIM-3, n=6) groups. At sacrifice, kidneys were harvested for morphology (glomerulosclerosis (GS), tubular injury and interstitial fibrosis), immunostaining (α-smooth muscle actin (α-SMA)) and platelet-derived growth factor receptor beta (PDGF-Rβ) and reverse transcriptase-polymerase chain reaction (RT-PCR) (MCP-1, ICAM-1, nephrin, and podocin) analysis.Results: Glomerulosclerosis, tubular injury and interstitial fibrosis in the simvastatin group was significantly lower than SN group (p<0.05). Simvastatin significantly reduced α-SMA expression (3.61±1.06 vs 7.91±1.26, p<0.05, SIM-1 vs SN; 2.86±0.61 vs 7.91±1.26, p<0.05, SIM-2 vs SN; 1.71±0.50 vs 7.91±1.26, p<0.05, SIM-3 vs SN), MCP-1 was markedly expressed in the 5/6 subtotal nephrectomy kidneys and was reduced with simvastatin (1.4±0.64 vs 0.57±0.23, p<0.05, SN vs SIM-1; 1.4±0.64 vs 0.6±0.26, p<0.05, SN vs SIM-2; 1.4±0.64 vs 0.52±0.21, SN vs SIM-3, p<0.05). Simvastatin did not increase nephrin expression, but it increased podocin expression significantly in the SIM-3 group.Conclusion: Simvastatin significantly attenuated GS, tubular injury and interstitial fibrosis through the downregulation of myofibroblast expansion and inflammatory mediators in mice with a 5/6 subtotal nephrectomy.
Excessive salt consumption is one of the hypertension factor leads to kidney disease, while telmisartan is one of antihypertensive drugs used in the therapy. Telmisartan not only blocks angiotensin receptor leads to the decrease of blood pressure, but it also activates peroxisome proliferator activated receptor gamma (PPAR-γ), inhibits transforming growth expression factor of beta-1 (TGFβ-1) and increases bone morphogenetic protein-7 (BMP-7). Whether telmisartan increases BMP-7 expression of excessive NaCl-induced Wistar rats are studied in this experiment. Twenty five male Wistars 2.5-3 months of age and 100-150 g BW rats were used in this research. They were grouped into 5, each consists of 5 rats. Group I (G I) as first negative control did not receive NaCl and telmisartan. G II as second negative control received NaCl but not telmisartan. G III, IV and V received NaCl and telmisartan 3, 6 and 12 mg/kg BW. The treatments were given every day within 8 weeks. At the day of 56 all rats were sacrificed by mean of neck dislocation and operated to take the kidney. The expression of BMP-7 was measured by immunohistochemistry technic. Data were expressed as mean ± standard deviation. They were analyzed by parametric (ANOVA) or nonparametric (Kruskal-Wallis) test. A value of p<0.05 was considered statistically significant. The results showed that intraglomerular and extraglomerular BMP-7 protein expression were higher in telmisartan-treated Wistar rats group than negative control group (p<0.05). In conclusion, intraglomerular and extraglomerular BMP-7 protein expression were higher in 8% sodium chloride-induced and telmisartan-treated male Wistar rats than the items of negative control group.
Garlic (Allium sativum L) is a medicinal plant traditionally used to relieve pain. Garlic's active constituents, allicin and triacremonone, have been proven to have antibacterial and antiinflammatory activity. The aim of this study was to investigate the effect of garlic ethanolic extract gel in gingival wound healing process of rats. Thirty male Wistar rats aged 10 weeks with body weight 200-250 g were subjected in this study. Rats were divided randomly into five groups with six rats in each group. Group I as negative control was given sodium carboxymethyl cellulose (Na CMC) base gel. Group II as positive control was given Benzydamine ® gel and Group IV-V were given garlic ethanolic extract gel at dose of 20, 40 and 80%, respectively. Each group was subdivided into two sub groups of three rats according to the decapitation period which were 5 th (D-5) and 7 th (D-7) day after the garlic extract gel application. Excisional wounds using punch biopsy, 2.5 mm in diameter, were created at the mandibular labial gingiva between right and left incisor teeth of the rats. The garlic extract gel of each preparation dose was then applied on the wound three times a day, starting at 0 day until 7 th day. The decapitation was conducted on the D-5 and D-7. Histological slides of wounded tissue were prepared. Epithelial thickness, new blood vessel, and number of fibroblast were examined. The results showed that the epithelial thickness of garlic ethanolic extract gel groups was significantly higher than control group (p<0.05), especially after 5 th day application. However, the number of new blood vessels and the amount of fibroblast of those groups were not significantly higher than control group (p>0.05). In conclusion, topical application of garlic ethanolic extract gel accelerates the gingival wound healing process in rats by increasing epithelial thickness. Kandungan aktif bawang putih yaitu alisin dan triakremonon terbukti mempunyai aktivitas antibakteri dan antiinflamasi. Penelitian ini bertujuan untuk mengkaji efek gel ekstrak etanol bawah putih pada proses penyembuhan luka gingiva tikus. Tiga puluh tikus jantan Wistar berumur 10 minggu dengan berat badan 200-250 g digunakan dalam penelitian ini. Tikus dibagi menjadi lima kelompok secara acak dengan enam ekor setiap kelompok. Kelompok I sebagai kontrol diberi natrium karboksilmetil selulosa (Na CMC). Kelompok II sebagai kontrol positif diberi gel Benzydamine ® . Kelompok III-IV diberi gel ektrak etanol bawang putih berturutturut pada dosis 20, 40 dan 80%.
