Pengembangan Domba Lokal melalui program Inseminasi Buatan memerlukan informasi mengenai kualitas semen, khususnya semen beku yang dalam pembuatannya memerlukan gliserol sebagai krioprotektan dengan kadar yang tepat. Penelitian ini bertujuan untuk mengetahui kualitas semen Domba Lokal dan mengetahui level gliserol yang menghasilkan kualitas semen post-thawing yang paling optimal. Penelitian dilakukan dengan menggunakan rancangan acak lengkap (RAL) dengan empat perlakuan level gliserol (4%, 5%, 6% dan 7%) dan 14 kali ulangan. Parameter yang diamati meliputi motilitas, abnormalitas dan recovery rate. Data yang diperoleh dianalisis menggunakan analisis ragam (ANAVA) dan perbedaan antar perlakuan diuji menggunakan Uji lanjut Duncan. Hasil penelitian menunjukkan bahwa level gliserol berpengaruh nyata (p<0.05) terhadap motilitas dan recovery rate namun tidak berpengaruh nyata terhadap abnormalitas sperma. Perlakuan level gliserol 5% nyata (p<0.05) menghasilkan motilitas (40,47%) dan recovery rate (46,53%) tertinggi dibanding perlakuan lainnya. Disimpulkan bahwa semen Domba Lokal memiliki kualitas yang baik dan memenuhi syarat untuk dilakukan pengolahan lebih lanjut sampai semen beku dengan level gliserol 5% menghasilkan kualitas semen post-thawing yang paling optimal.
Age is one of determinant factors on semen quality due to its relation with the development of male's reproductive tracts. Therefore, the objective of this study was to observe semen quality and viability at different level of ages in Javanese local sheep. The method used in this study was Complete Randomized Design with five groups of treatment consisted of groups of sheep aged: under 12 months (G1), 12-24 months (G2), 25-36 months (G3), 37-48 months (G4), and 49-72 months (G5). Each group consisted of four rams as replication and each replication was evaluated four times. Data collective which analyzed descriptively were: semen color, odor, viscosity, pH, sperm viability. While data on semen volume, sperm concentration, and sperm abnormality were analyzed using analysis of variance followed by Duncan's multiple range test for the differences among treatments. The result showed that no significant different found for semen quality parameters at different ages of ram (P>0.05), except for sperm motility (P<0.05). The lowest sperm motility (70.62%) found in G1 and significantly differs from other groups. The lowest sperm viability was also found in G1 after the chilled semen was kept for 4 to 5 days at 40% of motility, while the highest viability was in G3 by 9 days for 43.75% of motility. In conclusion: (1) Sperm motility of local ram semen was influenced by age; (2) Ram aged 25-36 months produces the highest motility of sperm and viability.
Aim of the research is to find out recovery rate and percentage spermatozoa X and Y after sexing with gradientt Percoll in Etawah Crossbreed Goat. Extender was used TRIS Egg Yolk. The semen sample from five male Etawah Crossbreed Goat were used in this research. Gradientt percoll performed 10 fraction layer with centrifugation time 10 minute at 2500 rpm. The data were analyzed descriptively. Result of the research shows that average recovery rate of spermatozoa X is 50.96±26.07 % and spermatozoa Y is 44.13±6.11 %. Average percentage of X sperm is 78.00±7.06 % and Y sperm is 76.10±5.95 % .
One of the estrus responses in livestock can be observed through the vulva, including through changes in the size of the vulva. This study aims to determine the effect of CIDR (Controlled Internal Drug Released) implant duration on changes in vulva size of Etawah Crossbreed goats. The study used an experimental method with a completely randomized design (CRD) with 3 types of treatment (P1 = CIDR implant for 10 days, P2 = CIDR implant for 12 days, P3 = CIDR implant for 14 days) with 6 replications. The parameters measured were the width and length of the vulva. The results of the descriptive analysis of the percentage of estrus showed that all livestock from various treatments experienced estrus. The results of the analysis of changes in the size of the vulvar cleft on day 1, day 2, and day 3 using the orthogonal polynomial test showed that there was no significant change on day 1 and day 2 between treatments, but there was a significant change on day-3. Based on the results of the study, it can be concluded that the length of the CIDR implant has a significant effect on changes in the size of the vulvar cleft.
