Background:
Conventional drugs like metronidazole for periodontitis has shown several drawbacks. So based on in-vitro and acute toxicity study of metronidazole loaded n-trimethyl quaternary ammonium chitosan nanoparticles (NTMET NPs), animal studies was carried out to proof enhancement of activities over periodontitis.
Objectives:
To assess the effect of nanoparticles through an intra-pocket drug delivery on body weight, food consumption, clinical signs, gingival index, tooth mobility, inflammatory changes in periodontal tissues, and serum analysis that leads into gingival damage, connective tissues and alveolar bone resorption.
Method:
P. gingivalis and ligature induced periodontitis model was used to evaluate the effect of drug in rats.
Results:
periodontitis was successfully induced by using P. gingivalis and ligature induced periodontitis model in rats. It reflect elevated gingival index, tooth mobility, inflammatory changes in tissues, IL-6, TNF- ɑ, and IL -1β when compared to control group and NTMET NPs treated groups. It was also observed that the damage caused to gingival tissues and resorption of alveolar bone was improved by using NTMET NPs.
Conclusion:
Based on present findings, we can conclude that NTMET NPs at a dose of 1.8 & 3.6 mg/ml has significant action on periodontal tissues and was able to reverse the effect of P. gingivalis as well as gingival damage and tooth mobility in rats at an administered dose, which are comparable to the IC50 value of a drug. The findings of the current study provide scientific preclinical data to support its clinical use for periodontitis via intra pocket drug delivery route.
Polyphenols from natural source are potential therapeutics that act alone or supplement anti-diabetic drugs in the prevention and treatment of diabetes. The present investigation was undertaken to study the effect of hydroalcoholic extract (HE) of fruits of Emblica officinalis on type 1 diabetic rats. Diabetes was induced by streptozotocin (STZ) (45 mg/kg i.v.). HE (100 mg/kg, p.o.) was administered for 4 weeks and at the end of treatment, blood samples were collected and analyzed for various biochemical parameters. STZ produced a diabetic state exhibiting all the cardinal symptoms such as loss of body weight, polydipsia, polyuria, glucosuria, polyphagia, hypoinsulinemia, and hyperglycemia associated with hypercholesterolemia and hypertriglyceridemia. Treatment with HE prevented cardinal symptoms and caused significant decrease in fasting serum glucose, AUCglucose, cholesterol, triglyceride, low-density lipoprotein (LDL) and very LDL in diabetic rats. However, insulin, AUCinsulin, and serum high-density lipoprotein level were not significantly altered by treatment. Treatment also reduced lipid peroxidation and increased anti-oxidant parameters in the liver homogenates of diabetic rats. Polyphenol enriched fraction of HE significantly improved disarranged carbohydrate and lipid metabolism of chemically induced diabetes in rats. The mechanism of its anti-diabetic activity appears to be either improvement in peripheral glucose utilization, increased insulin sensitivity, or anti-oxidant property.
:Congestive heart failure is, costly and deadly disease, is one of the leading causes of hospitalization and death worldwide. It is third most common cardio-vascular disease affect almost 2% of world population. Objective of present study is find out Cardioprotective and positive inotropic action of Seeds of Linum usitatissimum in Congestive Heart Failure. Methanolic extract and oil of seeds of Linum usitatissimum(MELU and OLU) were separated using soxhlet. Beneficial effects of methanolic extract and oil of seeds of Linum usitatissimum were carried out by doxorubicin (15mg/kg, i.p. within 3 weeks) to induce congestive heart failure in Wistar rat (invivo study). Following parameters like ECG recording, cardiac biomarkers like CPK-MB, LDH and SGOT and cytosolic Ca 2+ level were estimated. In doxorubicin induced congestive heart failure both MELU and OLU showed significant increased in cytosolic Ca 2+ level while significantly decreased QT interval as well as cardiac bio-markers. Present research work emphasizes that the seeds of Linum usitatissimum is beneficial in management of congestive heart failure.
