Extracellular vesicles (EVs), including exosomes and microvesicles, function in cell-to-cell communication through delivery of proteins, lipids and microRNAs to target cells via endocytosis and membrane fusion. These vesicles are enriched in ceramide, a sphingolipid associated with the promotion of cell senescence and apoptosis. We investigated the ceramide profile of serum exosomes from young (24–40 yrs.) and older (75–90 yrs.) women and young (6–10 yrs.) and older (25–30 yrs.) rhesus macaques to define the role of circulating ceramides in the aging process. EVs were isolated using size-exclusion chromatography. Proteomic analysis was used to validate known exosome markers from Exocarta and nanoparticle tracking analysis used to characterize particle size and concentration. Specific ceramide species were identified with lipidomic analysis. Results show a significant increase in the average amount of C24:1 ceramide in EVs from older women (15.4 pmol/sample) compared to those from younger women (3.8 pmol/sample). Results were similar in non-human primate serum samples with increased amounts of C24:1 ceramide (9.3 pmol/sample) in older monkeys compared to the younger monkeys (1.8 pmol/sample). In vitro studies showed that primary bone-derived mesenchymal stem cells (BMSCs) readily endocytose serum EVs, and serum EVs loaded with C24:1 ceramide can induce BMSC senescence. Elevated ceramide levels have been associated with poor cardiovascular health and memory impairment in older adults. Our data suggest that circulating EVs carrying C24:1 ceramide may contribute directly to cell non-autonomous aging.
Background: Fishing activity is a major source of marine litter on beaches, but most studies focus on recreational beaches. In this study, we assessed the abundance, composition, source and spatial distribution of marine litter on ten fishing beaches along Palk Bay, on the southeast coast of India. We also used social media to collect data on the impact of fishing litter on marine animals in the study area.
Results: A total of 4,227 marine litter and their average density of 0.70 items/m2 were collected. Plastic (84.54%) is the most abundant in the study area, followed by cloths (7.46%), glass (5.19%), and rubber (3.47%). From the result, the value of CCI ranges from 6.26 to 25.7 indicating that the coast is "moderately clean" to "extremely dirty". The majority of the marine litter collected originated from fishing activity. The most common are string and cord, fishing lines, fishing gear, floats and buoys.
Conclusion: The presence of this type of marine litter can produce serious environmental issues, such as ghost fishing, that impacts a large variety of marine species. A total of 25 of the olive ridley turtles (Lepidochelys olivacea) were entangled in fishing litter between 2018 and 2020. Most of the reported entanglements were due to discarded or lost fishing gear, which shows the importance of managing this type of litter in India. It is important to manage the litter related to the fishing activity by educating fishermen and organizing clean-up campaigns.
Myocarditis is an uncommon disease of adult and pediatric age group, characterized by inflammatory cell infiltrates, myocyte necrosis, or myocyte degeneration due to a myriad of causes and resulting in variable degrees of myocardial dysfunction. Which may be caused by infections, connective tissue disorders, granulomatous diseases, toxic agents or can be idiopathic. Viral infections are the most common etiology accounting for more than 50-60% of cases, particularly enterovirus (Coxsackie virus) and adenovirus groups. The disease is typically sporadic but may be epidemic. Manifestations of myocarditis range from asymptomatic or nonspecific generalized illness to acute cardiogenic shock and sudden death. In our case 10-year-old boy presented with history of fever and cough of 2 days duration. On examination patient found to be having tachycardia and tachypnea without other signs of heart failure. chest x-ray suggestive of bilateral pneumonia, ECG showed features of Myocarditis without any echocardiographic or cardiac enzyme abnormality. Child tested positive for corona virus rapid antigen test. Started on Favipiravir, corticosteroids and managed symptomatically. Child's heart rate normalized over a period of 4 days. Patient subsequently discharged with the advice of follow up echocardiography.
