The relationship between the structure and a property of a chemical compound is an essential concept in chemistry guiding, for example, drug design. Actually, however, we need economic considerations to fully understand the fate of drugs on the market. We are performing here for the first time the exploration of quantitative structure-economy relationships (QSER) for a large dataset of a commercial building block library of over 2.2 million chemicals. This investigation provided molecular statistics that shows that on average what we are paying for is the quantity of matter. On the other side, the influence of synthetic availability scores is also revealed. Finally, we are buying substances by looking at the molecular graphs or molecular formulas. Thus, those molecules that have a higher number of atoms look more attractive and are, on average, also more expensive. Our study shows how data binning could be used as an informative method when analyzing big data in chemistry.
Sestrins are highly conserved proteins that regulate cell growth, metabolism, survival
and proliferation under oxidative stress, genotoxic stress, hypoxia or endoplasmic
reticulum stress. Sestrins affect cell signaling by inhibiting the production of reactive
oxygen species, activating the AMP-activated protein kinase (AMPK), inhibiting
the mTOR pathway and acting as a positive regulator of autophagy. Therefore, their protective
role against cancer, metabolic disorders, cardiovascular diseases and neurodegeneration
is increasingly being postulated. The article describes the mechanisms of
action of sestrins and their meaning in aging and age-related diseases. The latest studies
indicating their physiological significance and role in key signaling pathways controlling
the cell metabolism and survival under stress conditions were also discussed.
The concept of ligand potency is briefly discussed here as well as
why this is still a challenge for its complete comprehension. In this
context, we explain also the meaning of ligand efficiency (LE), which has
been greeted with both enthusiasm and criticism among the drug design
audience. A full understanding of LE requires the complex interpretation of
the potency concept presenting the uncertainty similar to this of the Zeno
paradox. In reality, the efficiency of LE is caused by the high degree of
preference for slim pharma drug candidates.
