The agouti (Dasyprocta aguti Linnaeus, 1766) is a wild rodent belonging to the family Dasyproctidae that is found throughout Brazil and feeds on fruits and seeds. The aim of the present study was to describe the following features of the tongue of agouti: its morphological structures, the three-dimensional characteristics of the lingual papillae surface, the connective tissue cores (CTCs) and the epithelial cell ultrastructure. Four types of papillae were observed on the dorsal surface of the tongue with a triangular shape: filiform, fungiform, foliate and vallate. Filiform papillae were distributed throughout the tongue surface, and removal of the epithelial surface revealed conical CTCs and multifilaments. Fungiform papillae were observed in the rostral and middle regions, whereas foliate papillae developed in pairs on the lateral margin of the caudal region. Removal of the epithelium in these regions revealed CTCs with parallel laminar conformation. Vallate papillae were arranged in a V-shape in the caudal region, and their CTCs ranged in shape from elongate to ovoid. The ultrastructural components of the dorsal epithelium were the basal, spinous, granular and keratinised layers. A broad area with cytoplasmic projections was identified in the interface region between the lamina propria and the basal layer. Flattened cells with intermediate filaments were observed in the transitional region between spinous and granular layers. The keratinised layer was composed of superimposed epithelial cells where desmosomes and cell-surface microridges were observed. These structural features, including the three-dimensional aspects of the lingual papillae, the CTCs and the epithelial ultrastructure, indicate that when compared with other animals, particularly other rodent species, the morphological features of the tongue of agouti are relatively well developed, especially regarding foliate and vallate papillae.
The structural elements, tridimensional aspects of the lingual papillae, yours connective tissue cores (CTCs) and the ultrastructure of the dorsal epithelium surface of the tongue of guinea pig are examined. Filiform papillae and their CTCs lie throughout the tongue surface, featuring subtypes and different shapes. Fungiform papillae are dome-shaped, and their CTCs are few, with heterogenic disposition among the filiform papillae in the lingual apex and body and in grooves stretching to the side margin among the foliate papillae towards the lingual apex. Foliate papillae are well developed, and their CTCs are composed of five pairs of parallel seams limited by deep grooves placed bilaterally in the caudal region. Two vallate papillae are observed in the caudal region and reveal specialized structures which are developed and differentiated when they present compound elements by a double epithelial projection, in a column, similar to the shape of the foliate papilla. Further, the epithelium is organized by layers with ultrastructural components. Therefore, we conclude that the structural descriptions, dimensional aspects of tongue papillae, their CTCs and the ultrastructure of the tongue epithelium of guinea pigs are similar to other rodent mammals, however, with new descriptions as double epithelial projection constituting each vallate papilla.
BackgroundRenal cell carcinoma (RCC) is the most common type of kidney cancer, and represents the third most common urological malignancy. Despite the advent of targeted therapies for RCC and the improvement of the lifespan of patients, its cost-effectiveness restricted the therapeutic efficacy. In a recent report, we showed that synthetic phosphoethanolamine (Pho-s) has a broad antitumor activity on a variety of tumor cells and showed potent inhibitor effects on tumor progress in vivo.Methodology/Principal FindingsWe show that murine renal carcinoma (Renca) is more sensitive to Pho-s when compared to normal immortalized rat proximal tubule cells (IRPTC) and human umbilical vein endothelial cells (HUVEC). In vitro anti-angiogenic activity assays show that Pho-s inhibits endothelial cell proliferation, migration and tube formation. In addition, Pho-s has anti-proliferative effects on HUVEC by inducing a cell cycle arrest at the G2/M phase. It causes a decrease in cyclin D1 mRNA, VEGFR1 gene transcription and VEGFR1 receptor expression. Pho-s also induces nuclear fragmentation and affects the organization of the cytoskeleton through the disruption of actin filaments. Additionally, Pho-s induces apoptosis through the mitochondrial pathway. The putative therapeutic potential of Pho-s was validated in a renal carcinoma model, on which our remarkable in vivo results show that Pho-s potentially inhibits lung metastasis in nude mice, with a superior efficacy when compared to Sunitinib.Conclusions/SignificanceTaken together, our findings provide evidence that Pho-s is a compound that potently inhibits lung metastasis, suggesting that it is a promising novel candidate drug for future developments.
RESUMO.-O tipo de alimentação depende do ambiente em que o animal se encontra, sendo este um fator responsável pela alteração da morfologia, como a estratiϐicação e o nível de queratinização da língua, e a funcionalidade da mesma. Dentre as estruturas morfológicas funcionais da língua, as papilas vêm merecendo destaque devido a sua estreita relação com a dieta. Foram utilizadas duas espécies de cervídeos: cinco Mazama gouazoubira e duas Mazama americana, dividindo-se a língua em três partes: ápice, corpo e raiz. Two Cervidae species were used, ϐive Mazama gouazoubira and two Mazama americana. Their tongues were divided into three parts, apex, body and root, and comparatively analyzed by light and scanning electron microscopy. The ϐiliform, fungiform and vallate papillae were present in the two species' tongue and presented the same distribution, differing only in the quantity of vallate papillae on the root of the tongue, fact that might be related to the diet. Moreover, their distribution resembles that of other herbivore species.
Considering the physiological importance and need of greater morphophysiological knowledge of adrenal glands, the aims of present study were compare the morphometric data between left and right adrenal of male and female; perform a histological, scanning and transmission electron microscopy study showing tissue constitution of glands; finally, in order to define the presence and correct site of the cytochrome P450c17 expression in adrenal glands, immunohistochemical study of this enzyme was performed in 18 adrenal glands (right n=9 and left n=9) of nine adult Galea spixii (four males and five females). Right adrenal was more cranially positioned than left adrenal; dimensions (weight, length and width) of right adrenal was larger than left adrenal; no differences between male and female body and adrenal measurements were found; the morphology of cells and different amounts of lipid droplets may be related to the different demands of steroid hormones production, related to each zone of the adrenal cortex; and, the cytochrome P450c17 immunolocalization in fasciculate and reticular zone may be related with synthesis of 17-hydroxy-pregnenolone, 17-hydroxy-progesterone, dehydroepiandrosterone or androstenedione.Keywords: adrenal cortex, adrenal medulla, CYP, steroidogenic enzymes, wild rodents. Glândulas adrenais de preás (Galea spixii, Wagler, 1831):aspectos morfológicos e morfométricos ResumoConsiderando a importância fisiológica e a necessidade de melhor conhecimento morfofisiológico das glândulas adrenais, os objetivos do presente estudo foram comparar dados morfométricos entre adrenais direitas e esquerdas de machos e fêmeas; realizar estudos histológicos e microscopia eletrônica de varredura e transmissão revelando a constituição tecidual das glândulas; finalmente, foi realizado estudo imunohistoquímico com objetivo de definir a presença e correto sítio de expressão da citocromo P450c17 nas glândulas adrenais em 18 glândulas adrenais (direita n=9 e esquerda n=9) de nove Galea spixii adultos (quatro machos e cinco fêmeas).
