The brain of a 69-year-old man exhibited extensive granulomatous inflammation in the walls of arteries in the leptomeninges, associated with amyloid deposition in the media of the involved arteries. The extracranial arteries exhibited neither granulomatous inflammation nor amyloid deposition in their walls.
Our previous study showed that chronic exposure of the offspring of ICR/JCL mice to ferric nitrilotriacetate (Fe-NTA) , both the parents of which had been exposed to the same antigen, induced generalized amyloidosis in 69% of them. The amyloidotic offspring of 13 families obtained were designated as Fe-NTA-induced "F1 amyloidosis" mice. 16)The above study allows us a brief speculation that multiple injections of Fe-NTA stimulated chronically, and proliferated selectively F1 mouse clones to respond better to Fe-NTA than do their parents, eventually inducing macrophage dysfunction involved in amyloidogenesis in the F1 mice.In order to obtain further evidence for the above assumption the following studies comprising "fostera cre exneriments" were iindprta ken _
Materials and MethodsSix-week-old ICR/JCL mice (males weighing 28. 2 to 31. 5g, females weighing 23. 8 to 25. 8g)were injected intraperitoneally with ferric nitrilotriacetate (Fe-NTA) O. 5 to 1. 0mg/100g body weight, once a day, 6 times per week (Awai et al. , 1979 2)).Group I.The offspring from reciprocal crossing between 3 females which had previously received (a) 99 injections of Fe-NTA over 122 days, (b) 95 injections over 112 days or (c) 95 injections over 112 days and 3 untreated males, respectively, were administered from the 6th week after birth 150 to 256 injections of Fe-NTA over a period of 180 to 300 days. Offspring used : (a) 10 (3 males and 7 females), (b) 11 (6 males and 5 females), (c) 10 (3 males and 7 females).Group II.Offspring from reciprocal crossing between parental mice, both of which had previously received days from the first week after completion of lactation by an untreated normal puerperal wet nurse. Offspring used : (a) 9 (2 males and 7 females), (b) 9 (2 males and 7 females) , (c) 10(5 males and 5 females), (d) 11 (2 males and 9 females) , (e) 9 (3 males and 6 females).Group El.Offspring from reciprocal crossing between untreated males and females (females were used as puerperal wet nurses in Group II) were administered 189 to 256 injections of Fe-NTA over a period of 221 to 300 days from the first week after completion of lactation by puerperal wet days (The offspring from the above puerperal wet nurses were used as fosterling in Group ).Offspring used : (a) 9 (1 male and 8 females), (b) 9 (2 males and 7 females), (c) 9 (3 males and 6 females), (d) 9 (4 males and 5 females), (e) 9 (2 males and 7 females) . (a) Of 9 offspring 3 (1 male and 2 females) revealed generalized amyloidosis at an incidence of about 33%.(b) Of 9 offspring 4 (females) revealed generalized amyloidosis at an incidence of about 44%.(c) Of 10 offspring 3 (2 males and 1 female) revealed generalized amyloidosis at an incidence S -45 of 30%.(d) Of 11 offspring 3 (2 males and 1 female) revealed generalized amyloidosis at an incidence of about 27%.(e) Of 9 offspring 6 (2 males and 4 females) revealed generalized amyloidosis at an incidence of about 67%. Moreover, amyloidotic mice in groups I and II frequently revealed characteristic widespread erosions localize...
