This investigation was conducted to study the range of TDZ-induced morphogenetic responses displayed by foliar explants of Kalanchoe pinnata with or without an epiphyllous bud and to assess their possible auxin mediation by using potent anti-auxin TIBA. Each epiphyllous bud developed into a healthy plantlet when cultured in vitro without nutrient/hormonal supplements. The bud on 10 )6 M TDZ-treated leaf disc produced a compact, dwarf and rootless plantlet having achlorophyllous leaves with increased number of marginal notches. TDZ also caused a significant enlargement of the leaf disc as well as induced an organized hypertrophic growth at a specific site, with either effect being more pronounced in discs without an epiphyllous bud. Treatment with 10 )4 M TIBA completely checked the plantlet growth. In combined dispensations, the TDZ-induced responses were modified by the anti-auxin TIBA in more than one way. Whereas it caused a complete reversal of hypertrophic growth on leaf discs and partially restored pigmentation of plantlet leaves, it could not annul the leaf disc enlargement. The diverse morphogenetic modulations were tissue and response specific. A complete reversal of TDZ-induced leaf disc hypertrophy by TIBA indicated an auxin mediation, whereas a complete non-reversal of the induced leaf disc enlargement pointed to the noninvolvement of auxin, for the respective responses. The latter response could probably be manifested through cytokinin-activity of TDZ, while still others may not be interpreted exclusively through either one of the two hormonal options.
India has a large number of paper manufacturing units, with mills varying in size of operations and type of product, scattered all over the length and breadth of the country. Present study deals with the survey of four different pulp and paper mills (Mill A, Mill B, Mill C and Mill D) in India, differing in their scale of operation as well as raw material usage. Physico-chemical characterization of wastewaters produced by different processing units like digestor house liquors, bleaching section, paper machine backwaters, combined inlet to the ETP and outlet to ETP, for all the four mills, was conducted. Different parameters like pH, Color, COD, BOD, percent Na, SAR, TDS and Total hardness were analyzed and compared. Highly significant differences were thus found between all the processing units of all four mills with p values < 0.01. This evaluation showed that though these mills have effluent treatment facilities installed in their premises, improvements are still needed to increase their efficiencies for treating the discharged wastewaters.
Thidiazuron induces a variety of effects in foliar explants of K. pinnata cultured in vitro. Of these, the induction of an organized hypertrophic growth at the vein ending on the proximal side of leaf disks is a significant morphogenetic effect. This polarized hypertrophy is considered an exclusive auxin-mediated response, as the effect is completely reversed by auxin efflux inhibitor 2,3,5-triiodobenzoic acid. With the magnitude of hypertrophic growth from individual disks taken as a manifestation of putative auxin levels, the occurrence of a gross, decreasing bioactive auxin gradient from median-basal to peripheral locations across the leaf plane has been observed.
