Abortion is the most common and controversial issue in many parts of the world. Approximately 46 million abortions are performed worldwide every year. The world ratio is 26 induced abortions per 100 known pregnancies. Pakistan has an estimated abortion rate of 29 abortions per 1,000 women of reproductive age, despite the procedure being illegal except to save a woman's life. 890,000 abortions are performed annually in Pakistan. Many government and non-government organizations are working on the issue of abortion. Muslim jurists are unanimous in declaring that after the fetus is completely formed and has been given a soul, abortion is haram (forbidden).
ABSTRACT. Micro-RNAs are cellular components regulating gene expression at the post-transcription level. In the present study, artificial micro-RNAs were used to decrease the transcript level of two genes, AtExpA8 (encoding an expansin) and AHL25 (encoding an AT-hook motif nuclear localized protein) in Arabidopsis thaliana. The backbone of the Arabidopsis endogenous MIR319a micro-RNA was used in a site-directed mutagenesis approach for the generation of artificial micro-RNAs targeting two genes. The recombinant cassettes were expressed under the control of the CaMV 35S promoter in individual A. thaliana plants. Transgenic lines of the third generation were tested by isolating total RNA and by subsequent cDNA synthesis using oligo-dT18 primers and mRNAs as templates. The expression of the two target genes was checked through quantitative realtime polymerase chain reaction to confirm reduced transcript levels for AtExpA8 and AHL25. Downregulation of AtExpA8 resulted in the formation of short hypocotyls compared with those of the wild-type control in response to low pH and high salt concentration. This technology could be used to prevent the expression of exogenous and invading genes posing a threat to the normal cellular physiology of the host plant.
Water is one of the necessary ingredients for construction materials. Billions of gallons of clean water are wasted during the development of fired clay bricks. Similarly, the waste of clean water is a global issue. In this study, we develop fired clay bricks with the help of wastewater for the first time and compare these with clay bricks produced using groundwater, which is the conventional method. Both destructive (i.e., compressive strength (CS)) and non-destructive (i.e., ultrasonic pulse velocity (UPV)) tests are conducted on all fired clay brick specimens as per the American Society for Testing and Materials (ASTM). Physical (i.e., dimensions) and durability (water absorption, efflorescence, etc.) tests are also conducted. All kinds of brick satisfied the standard requirements of physical and durability characteristics. Similar or better strength of bricks were achieved using wastewater. The study concludes that the testing results of wastewater bricks were significantly 15–25% higher compared with groundwater-fired clay bricks. A large amount of wastewater can be used to develop bricks, and clean water can be saved to attain circular economy goals. Therefore, this study will help not only in developing low-cost bricks but also in saving clean water.
Cardiovascular diseases (CVD) are the leading cause of death all over the world. Beside general risk factors, there are some genetic factors which lead to cardiovascular diseases. Various nuclear DNA mutation and also mitochondrial DNA mutations have been related with cardiovascular diseases. In the present study, a total of 21 samples were collected from different families residing in district Dir. DNA was extracted from buccal epithelial cells using saliva. The mitochondrial tRNA leu (MT TL1) gene was amplified by PCR and 10 samples of different families were sequenced. The sequence was aligned with revised Cambridge Reference Sequence (rCRS) accession # NC-012920.1. It is concluded that cardiovascular diseases in our subjects are not due to mutation in the mitochondrial leucine tRNA gene. However, a large population of subjects with cardiovascular diseases needs to be studied and whole mitochondrial DNA is needed to be sequenced in the subjects with CVD. This will give an idea about the probable DNA marker which can be used to prevent loses due to these diseases at a very early stages.
