In this study, the effects of above and below-ground extracts of Centranthus longiflorus subsp. longiflorus plant, commonly found in Turkey, on antioxidant, antimicrobial and DNA damage were evaluated. Plant extracts were prepared by applying three different solvents (hexane, methanol and ethanol). The antimicrobial activity tests of the extracts were performed using four different standard strains and one yeast. DPPH, total phenolic content calculation and CUPRAC methods were applied for antioxidant activity studies. Additionally, the effects of plant extracts on DNA damage were investigated using pBR322 plasmid DNA. According to the data obtained, especially the below-ground hexane (MIC value:375µg/mL) extract showed more antimicrobial activity than other plant extracts, and it was found to be more effective Gram negative bacteria. The highest antioxidant activity was determined in extracts prepared with above (IC50 value of methanol extract:4.5mg/mL) and below-ground (IC50 value of methanol extract:5.7mg/mL) methanol. The above (93,9 µg GAE/mL) and below-ground (96.9 µg GAE/mL) methanol extracts were seen to have high total phenolic content. It has also been observed that above-ground hexane and methanol extracts have no effect on pBR322 plasmid DNA, but other extracts affect pBR322 plasmid DNA in the direction of degradation or deformation. Especially, the extracts of the above and below-ground ethanol had the effect of completely eliminating the open ring form. Therefore, it was concluded that this taxon could be widely used in the treatment and prevention of oxidative stress-related diseases in the future.
Bu makalede, Heliotropium suaveolens ve Heliotropium myosotoides’in farklı organlarından elde edilen ekstraktların antioksidan, antimikrobiyal aktiviteleri ve DNA hasarı üzerine etkileri değerlendirildi. H. suaveolens’in vejetatif ve generatif gelişme periyotlarındaki farklı organlarına, H. myosotoides’in generatif ve vejetatif gelişme periyodundaki topraküstü ve toprakaltı organlarına hekzan, etanol ve etil asetat uygulanmıştır. Daha sonra bitki ekstrakları DMSO içerisinde çözülerek antioksidan, antimikrobiyal ve plazmit DNA çalışmalarında kullanılmıştır. H. suaveolens’in vejetatif gelişme periyodunda yaprak, generatif gelişme periyodunda ise yaprak ve çiçek etanol ekstraktlarında yüksek antioksidan aktivite görülmüştür. Ancak H. suaveolens’in vejetatif gelişme periyodunda gövde, generatif gelişme periyodunda kök ve gövde etanol ekstraklarının zayıf antioksidan aktivite gösterdiği tespit edilmiştir. H. suaveolens’in hekzan ekstraktlarında ise antioksidan aktivite vejetatif ve generatif gelişme periyotlarındaki bütün organlarında bulunmuştur. H. myosotoides'in generatif büyüme periyodundaki topraküstü ve toprakaltı organlarının etanol ve hekzan ekstraktlarının zayıf antioksidan aktiviteye sahip olduğu görülmüştür. Ancak bu iki türün etanol, hekzan ve etil asetat ekstraktlarının antimikrobiyal aktivite ve DNA hasarı göstermediği belirlenmiştir. H. suaveolens’in antioksidan aktivite gösteren organlarının parfümeri ve kozmetik sanayisinde değişik preparatların hazırlanmasında ve içerdiği glikozitlerden dolayı bal üreticileri tarafından kullanılabileceği önerilmiştir.
Iris taochia is an elegant endemic plant in Türkiye and it has limited distribution. In this study, cytotoxic effects of ethanolic extracts from different parts and concentrations extracted from I. taochia collected from the surroundings of Tortum (Erzurum), on A549 human lung adenocarcinoma cell line were investigated. Cytotoxicity of extracts were evaluated by MTT (3-(4,5-Dimethylthiazol-2-yl)-2,5-Diphenyltetrazolium Bromide) method. Apopototic activity of IC50 values of extracts were evaluated with Annexin V and Caspase 3/7 assays. Ultrastructural changes of IC50 doses treated cells were investigated by transmission electron microscopy. As a result, it was determined that ethanol extract of I. taochia showed significant cytotoxic activity on A549 cells after 24 hours the extract a dose-dependent reduction in cell viability. IC50 values of above and below ground parts ethanolic extracts were determined as 7 µg/ml and 20 µg/ml respectively. Specifically, apoptosis inducing effect was increased at 7 and 20 µg/ml concentrations by 24 hours. We found that I. taochia ethanol extracts had antiproliferative and apoptotic effects on the human lung adenocarcinoma cells A549. However, further studies at molecular level are required to support our findings and to elucidate chemoproteventive and chemotherapeutic effects of I. taochia on lung cancer.
