Premature fracture of an axle under torsional load occurred after a tracked military tank had experienced field testing for only 80 kilometers. Visual metallographic examinations were performed with optical microscope (OM) and scanning electron microscope (SEM). The investigation demonstrates that the premature fracture is caused by metallurgical problems inside the axle where the primary and secondary cracks originate, propagate, and eventually result in final catastrophic rupture through torsional fatigue. The failure mechanism is summarized and improvement of the fatigue lifetime for the axle is recommended.
This research first analyzes the features of sports network information resources, and further illustrates the existing problems on current research information resources gaining from sports network academic information resources, theses problems mainly includes less information accuracy, uneven quality of information and rapidly update of academic information. Meanwhile the research makes focus on standards on sports academic workers gaining sports network information resources, i.e. focus on information accuracy, enforce academic value, strengthen the credibility of information releaser, pay attention on information stability and the extent of utility, at last, it points out the standards on evaluation to sports network information resources, the standards includes accuracy of the information, the information authority, the breadth and depth of information, the purpose of providing information, information of the objectivity and timeliness of information. The author hope that this research could provide valuable referring suggestions to sports workers on gaining network information resources.
Protein modifications such as post-translational modifications (PTMs) and sequence variants (SVs) occur frequently during protein biosynthesis and have received great attention by biopharma industry and regulatory agencies. In this study, an aberrant peak near light chain (LC) was observed in the non-reduced capillary electrophoresis sodium dodecyl sulfate (nrCE-SDS) electrophoretogram during cell line development of one bispecific antibody (BsAb) product, and the detected mass was about 944 Da higher than LC. The corresponding peak was then enriched by denaturing size-exclusion chromatography (SEC-HPLC) and further characterized by nrCE-SDS and peptide mapping analyses. De novo mass spectra/mass spectra (MS/MS) analysis revealed that the aberrant peak was LC related sequence variant, with the truncated C-terminal sequence “SFNR” (“GEC”deleted) linked with downstream SV40 promotor sequence “EAEAASASELFQ”. The unusual sequence was further confirmed by comparing with the direct synthetic peptide “SFNREAEAASASELFQ”. It was demonstrated by mRNA sequencing of the cell pool that the sequence variant was caused by aberrant splicing at the transcription step. The prepared product containing this extension variant maintained well-folded structure and good functional properties though the LC/Heavy chain (HC) inter-chain disulfide was not formed. Several control strategies to mitigate the risk of this LC related sequence variant were also proposed.
