ABSTRACT Cholesterol is a fat that is needed by the body, but if it is in excess, it can cause accumulation of cholesterol in the walls of blood vessels so that it can slowly harden the walls of blood vessels and then inhibit blood flow and cause atherosclerosis. To know the cholesterol level in the blood, it is necessary to check the cholesterol using blood serum and blood plasma samples. However, it is advisable to use blood serum to check cholesterol so that anticoagulant contaminants do not occur. To obtain blood serum can be done by the centrifuge method and not centrifuged (precipitated). Cholesterol examination using the centrifuge method is directly examined without delay, while the non-centrifuge method is subject to a maximum delay of 2 hours after sampling. This study aims to determine whether there is a difference in total blood cholesterol levels in centrifuged and non-centrifuged serum at the Ratnasari Medical Center Clinic in March-May 2020. The results showed that the average cholesterol value for centrifuged samples was 170.1 mg / dl and not. centrifuged at 168.8 mg / dl. Based on the Paired t-test, the p-value was 0.07 (p> 0.05), which means that there was no significant difference between centrifuged and non-centrifuged serum (p> 0.05). The conclusion of this study is that there is no difference in the value of total cholesterol levels in centrifuged and non-centrifuged serum at the Ratnasari Medical Center Clinic Rawamangun. Keywords: Cholesterol, Blood Serum, Centrifuge, Not Centrifuged ABSTRAK Kolesterol merupakan suatu lemak yang dibutuhkan oleh tubuh tetapi jika jumlahnya berlebih dapat menyebabkan penimbunan kolesterol di dinding pembuluh darah sehingga secara perlahan dapat mengeraskan dinding pembuluh darah kemudian menghambat aliran darah dan menyebabkan aterosklerosis. Untuk mengetahui kadar kolesterol di dalam darah diperlukan pemeriksaan kolesterol menggunakan sampel serum darah dan plasma darah. Namun, pemeriksaan kolesterol lebih dianjurkan menggunakan serum darah agar tidak terjadi kontaminan oleh antikoagulan. Untuk memperoleh serum darah dapat dilakukan dengan metode disentrifus dan tidak disentrifus (diendapkan). Pemeriksaan kolesterol menggunakan metode disentrifus langsung dilakukan pemeriksaan tanpa ditunda sedangkan metode tidak disentrifus dilakukan penundaan pemeriksaan maksimal 2 jam setelah pengambilan sampel. Penelitian ini bertujuan untuk mengetahui ada tidaknya perbedaan kadar kolesterol total darah pada serum disentrifus dan tidak disentrifus di Klinik Ratnasari Medical Centre pada bulan Maret-Mei 2020. Hasil penelitian didapatkan nilai rata-rata kolesterol untuk sampel disentrifus sebesar 170,1 mg/dl dan tidak disentrifus sebesar 168,8 mg/dl. Berdasarkan Uji Paired t-test didapatkan hasil p-value 0.07 (p>0.05) yang berarti tidak terdapat perbedaan bermakna antara serum disentrifus dan tidak disentrifus (p > 0.05). Kesimpulan penelitian ini adalah tidak terdapat perbedaan nilai kadar kolesterol total pada serum yang disentrifus dan tidak disentrifus di Klinik Ratnasari Medical Centre Rawamangun.
