Cryoprotectants are known to have protective effects against cryodamage to spermatozoa. In this study, the cryoprotective effects of two cryoprotectants (glycerol, ethylene glycol) and cryoprotectants/trehalose combinations on frozen-thawed ram spermatozoa were investigated at the ultrastructural level. For this purpose, ejaculates collected from Konya Merino rams were pooled and diluted with a tris-based extender containing additives, including 5% glycerol, 3% glycerol +60 mM trehalose, 1.5% glycerol +100 mM trehalose, 5% ethylene glycol, 3% ethylene glycol +60 mM trehalose, and 1.5% ethylene glycol +100 mM trehalose. They were all cooled to 5°C and then frozen in 0.25 mL French straws in liquid nitrogen. The samples were thawed at 37°C and centrifuged to remove the diluents. Then, they were processed using a scanning transmission electron microscope. In the statistical analysis, the number of ultrastructurally cryodamaged and intact spermatozoa were counted in longitudinal and transverse ultrathin sections in all groups by electron microscopic examination. The amount of intact spermatozoa in the groups containing 5% ethylene glycol and 1.5% ethylene glycol +100 mM trehalose was found to be higher than other groups (p < 0.05). As a result, it was suggested that the groups of 5% ethylene glycol and 1.5% ethylene glycol +100 mM trehalose provided the highest protection for the ultrastructural morphology of frozen-thawed Konya Merino ram spermatozoa among the groups.
Abstract. The aim of this study was to evaluate the effects of premating ewe weight and age on ewe reproductive performance, lamb growth and survival in Central Anatolian Merino (CAM) sheep.The trial included 4935 ewes and 5548 lambs from 23 breeder flocks in Eskişehir Province. The live weights (LWs) and ages of the ewes were recorded 1 week before the mating season. The number of lambs born, fertility rate, multiple birth rate (MBR), and lamb survival rate (SR) at days 75 and 120 were determined. The effects of the LW and age of the ewe were found to be important for the reproductive performance of ewes and for the lamb's growth rate (P < 0.05-0.001). Ewe MBRs and the birth weight (BW) and LW of the lambs at days 75 and 120 increased proportionally with the increase in LW in ewes prior to mating. The fertility rate and MBR were lower (P < 0.05-0.001) in 1.5-year-old ewes than in older ewes. However, lambs' SR at days 75 and 120 were not affected by the premating LW and age of the ewe. In summary, a better reproductive performance and lamb growth rate in CAM flocks may be achieved by the selection of breeding ewes from ewes weighing more than 60 kg.
The aim of this study was to evaluate the effects of the transfer side, transfer location, cervix transfer score, type and diameter of corpus luteum (CL) during embryo transfer on pregnancy rates in beef heifers. Progesterone‐based synchronization and superovulation protocol were applied to Simmental cows used as donors (n = 168). Uterine flushings were performed on day 7 following artificial insemination. Obtained Code I (excellent or good) and II (fair) quality embryos were transferred to recipient beef heifers (n = 561). During embryo transfer, side of transfer (right or left), transfer location (the cranial or middle third of uterine horn), cervix transfer score (easy, moderate or difficult) and type (CLa, CLb and CLc) and diameter of CL were determined. Pregnancy rates following the transfer of Code I and II embryos were 44.66% and 33.07%, respectively (p < .05). The rates of pregnancy after transfers to the right and left uterine horn were 37% and 42.2%, respectively (p > .05). The pregnancy rates were 41.2%, 34.9% and 30.3% for cervix transfer scores as easy, moderate and difficult, respectively (p > .05). Pregnancy rates after transfer to the cranial third and middle third were 41.06% and 29.67%, respectively (p < .05). According to types of CL, pregnancy rates were 31.7%, 40.4% and 45.3% for CLa, CLb and CLc, respectively (p < .05). Moreover, it was found that as the CL diameter increased, the pregnancy rates increased. As a result, it was concluded that there was no effect of side of transfer and cervix transfer score, but embryo quality, transfer location, type and diameter of CL had significant effects on the pregnancy rate during embryo transfer in beef heifers.
